Real-time 2P billeddannelse af intakt væv er forpligtet til at undersøge NPC kinetik og interaktioner efter transplantation ind i demyeliniserede mus rygmarven. Musemodeller for demyelination, herunder eksperimentel autoimmun encephalomyelitis (EAE) og intrakraniel infektion med neuroadapted mus hepatitis virus (MHV), er fremragende værktøjer til at studere molekylære veje og cellulære interaktioner forbundet med sygdommen. De har ført til og støttet effektiviteten af FDA godkendte farmaceutiske behandlinger, primært rettet mod ophør af autoimmunitet og inflammation 1. Men når endogene remyelinering mislykkedes, de aktuelt godkendte behandlinger ikke effektivt reparere demyeliniserede læsioner i centralnervesystemet. For at omgå disse problemer og undersøgelse celle-celle-interaktioner inden læsioner langs den ventrale rygmarv har vi udviklet en simpel ex vivo 2P billeddannelse fremstilling 6. Denne undersøgelse er en opfølgning af en tidligere metoder publikation, som visteproceduren til omplantning forstærket grønt fluorescerende protein (eGFP) som udviser NPC i rygmarven af mus efter JHMV stamme af MHV-induceret demyelinisering 14. Fem uger gamle mus inficeret med JHMV og transplanteret med eGFP-NPC&#39;ere på thorax niveau 9 på dag 14 efter infektion. two-photon imaging of cellular dynamics in the mouse spinal cord

Two-photon (2P) microscopy is utilized to reveal cellular dynamics and interactions deep within living, intact tissues. Here, we present a method for live-cell imaging in the murine spinal cord. This technique is uniquely suited to analyze neural precursor cell (NPC) dynamics following transplantation into spinal cords undergoing neuroinflammatory demyelinating disorders. NPCs migrate to sites of axonal damage, proliferate, differentiate into oligodendrocytes, and participate in direct remyelination. NPCs are thereby a promising therapeutic treatment to ameliorate chronic demyelinating diseases. Because transplanted NPCs migrate to the damaged areas on the ventral side of the spinal cord, traditional intravital 2P imaging is impossible, and only information on static interactions was previously available using histochemical staining approaches. Although this method was generated to image transplanted NPCs in the ventral spinal cord, it can be applied to numerous studies of transplanted and endogenous cells throughout the entire spinal cord. In this article, we demonstrate the preparation and imaging of a spinal cord with enhanced yellow fluorescent protein-expressing axons and enhanced green fluorescent protein-expressing transplanted NPCs.
