En temps réel 2P imagerie du tissu intact est nécessaire pour enquêter sur la cinétique et interactions NPC après la transplantation dans la moelle épinière de souris démyélinisée. Imagerie intravitale 2P est couramment utilisé pour déterminer la dynamique cellulaire sur la face dorsale de la moelle épinière chez les souris vivant, et a été utilisé pour étudier dorsale démyélinisation maladie démyélinisante 17-19. Toutefois, en raison des PNJ transplantées migrent à la matière blanche...

Des modèles murins de démyélinisation, y compris l&#39;encéphalomyélite auto-immune expérimentale (EAE) et l&#39;infection par le virus intracrânienne neuroadapted de l&#39;hépatite de la souris (MHV), sont d&#39;excellents outils pour étudier les voies moléculaires et interactions cellulaires associés à la maladie. Ils ont mené et soutenu l&#39;efficacité des thérapies pharmaceutiques approuvé par la FDA, ciblant principalement la cessation de l&#39;auto-immunité et l&#39;inflammation 1. Cependant, une fois la remyélinisation endogène a échoué, les thérapies actuellement approuvés ne réparent pas efficacement les lésions démyélinisées dans le système nerveux central. Par conséquent, les thérapies de réparation axé à ce stade de la maladie sont décisifs pour le soulagement des symptômes chroniques et l&#39;amélioration de la qualité de vie. Récemment, des cellules précurseurs neurales (PNJ) sont venus à l&#39;avant-garde comme une modalité thérapeutique régénérative potentiel pour cibler des zones d&#39;inflammation et de démyélinisation. Plusieurs études ont mis en évidence la capacité des PNJ pour induire endógenonous remyélinisation et participer directement à la remyélinisation 2-8. Parce que les PNJ sont impliqués dans la remyélinisation directe, il est impératif de comprendre leur cinétique et les interactions avec des cellules endogènes après la transplantation. Après la transplantation, NPC migrent ventralement à des zones de blanc atteinte de la substance, puis rostrale et caudale par rapport au site de transplantation 5,9. La cinétique de la migration diffèrent en réponse aux signaux environnementaux; NPCs transplantées dans la moelle épinière non endommagés ont de plus grandes vitesses que PNJ transplantées dans la moelle épinière endommagée 6. Après une période de migration, transférés PNJ prolifèrent abondamment, à un taux supérieur dans une moelle épinière endommagée par rapport à une moelle épinière intacte 6. Enfin, la majorité des PNJ se différencier en oligodendrocytes et initier 4,6,9 de la remyélinisation directe. La lésion démyélinisée est complexe et peut comprendre une population diverse de cellules à divers stades d&#39;unctivation. Par exemple, une lésion active la sclérose en plaques (MS) peut comprendre une importante population de cellules T activées, la microglie et des macrophages M1 M1, mais une lésion chronique silencieux MS peut être composée essentiellement d&#39;astrocytes réactifs avec quelques cellules inflammatoires 10-13. En raison de la diversité des cellules effectrices, à deux photons (2P) imagerie dans des modèles murins de démyélinisation est un outil extrêmement utile pour aider à comprendre les interactions cellulaires locales au sein de la lésion. Dans la SEP et de nombreux modèles de recherche de MS largement utilisées, la majorité des lésions sont situées sur la face ventrale de la moelle épinière, une région inaccessible à l&#39;imagerie intravitale 2P en raison de la profondeur de la lésion et la forte teneur en lipides de la moelle épinière. Pour contourner ces problèmes et interactions étude cellule-cellule au sein de lésions long de la moelle épinière ventrale nous avons développé simple vivo 2P préparation d&#39;imagerie 6 ex. Cette étude fait suite à une précédente publication méthodes, qui a montréla procédure de transplantation protéine fluorescente verte améliorée (eGFP) PNJ dites-vous dans la moelle épinière de souris après souche JHMV de démyélinisation MHV-induite 14. Souris âgées de cinq semaines sont infectées par JHMV et transplantées avec eGFP-PNJ au niveau thoracique 9 sur 14 jours post-infection. Le protocole présenté ici fournit des instructions détaillées sur la façon d&#39;extraire la moelle épinière, faire une préparation ex vivo agarose et image transplanté interactions eGFP-NPC avec la protéine fluorescente jaune améliorée (EYFP) des axones dites-vous. Les souris exprimant sous la EYFP Thy1 promoteur spécifique de neurones ont été utilisés dans cette procédure 15. Seuls quelques-uns des axones exprimer EYFP, le rendant utile pour l&#39;imagerie axones individuels. Ici, nous montrons la moelle épinière prélevés à sept jours post-transplantation; Toutefois, la moelle épinière peuvent être extraites à tout point de temps après la transplantation. Bien que nous montrons interactions avec des PNJ axones endommagés, notre protocole peut être utilisé en combinaison avec des marqueurs fluorescents génétiquesd&#39;autres types de cellules à examiner une multitude d&#39;interactions cellulaires qui se produisent tout au long de la moelle épinière de souris.

two-photon imaging of cellular dynamics in the mouse spinal cord

Two-photon (2P) microscopy is utilized to reveal cellular dynamics and interactions deep within living, intact tissues. Here, we present a method for live-cell imaging in the murine spinal cord. This technique is uniquely suited to analyze neural precursor cell (NPC) dynamics following transplantation into spinal cords undergoing neuroinflammatory demyelinating disorders. NPCs migrate to sites of axonal damage, proliferate, differentiate into oligodendrocytes, and participate in direct remyelination. NPCs are thereby a promising therapeutic treatment to ameliorate chronic demyelinating diseases. Because transplanted NPCs migrate to the damaged areas on the ventral side of the spinal cord, traditional intravital 2P imaging is impossible, and only information on static interactions was previously available using histochemical staining approaches. Although this method was generated to image transplanted NPCs in the ventral spinal cord, it can be applied to numerous studies of transplanted and endogenous cells throughout the entire spinal cord. In this article, we demonstrate the preparation and imaging of a spinal cord with enhanced yellow fluorescent protein-expressing axons and enhanced green fluorescent protein-expressing transplanted NPCs.
