Protein variants have been shown to be involved in the progression of many neurodegenerative diseases such as Alzheimer&#8217;s, Parkinson&#8217;s, ALS and FTD1,2,3,4,5,6,7,8,9,10,11. Aquí ampliar este proceso para permitir la generación de reactivos que se unen selectivamente a la proteína específica de la enfermedad variantes directamente a partir de muestras de tejidos humanos. En estudios futuros se pretende investigar más a fondo no sólo el valor de diagnóstico de los reactivos aislados aquí, sino también evaluar su relevancia terapéutica para el tratamiento de ALS. En general, nuestra nueva tecnología basada biopanning AFM debe ser aplicable al aislamiento de cualquier variante de proteína específica de la enfermedad en cualquier material biológico sin la necesidad de purificación de proteínas o modificación, incluso cuando el antígeno diana concentrations son extremadamente bajos.

novel atomic force microscopy based biopanning for isolation of morphology specific reagents against tdp-43 variants in amyotrophic lateral sclerosis

Debido variantes de proteínas juegan un papel crítico en muchas enfermedades incluyendo TDP-43 en la esclerosis lateral amiotrófica (ELA), la alfa-sinucleína en la enfermedad y el beta-amiloide y tau en la enfermedad de Alzheimer, es de vital importancia para el desarrollo de reactivos específicos morfología que pueden dirigirse selectivamente Parkinson estas proteínas específica de la enfermedad variantes para estudiar el papel de estas variantes en la patología de la enfermedad y para posibles aplicaciones diagnósticas y terapéuticas. Hemos desarrollado nuevos microscopía de fuerza atómica (AFM) técnicas biopanning base que permiten el aislamiento de los reactivos que reconocen selectivamente variantes de proteínas específicas de la enfermedad. Hay dos fases clave implicados en el proceso, las fases de panning negativos y positivos. Durante la fase de panning negativo, los fagos que son reactivos a los antígenos fuera de objetivo se eliminan a través de múltiples rondas de cribado sustractiva utilizando una serie de antígenos cuidadosamente seleccionados fuera de objetivo. Una característica clave de la negativafase de la panorámica es la utilización de imágenes de AFM para vigilar el proceso y confirmar que se eliminan todas las partículas de fago no deseados. Para la fase de panning positivo, el antígeno diana de interés se fija sobre una superficie de mica y fagos unidos se eluyen y se tamiza para identificar los fagos que se unen selectivamente al antígeno diana. La variante de proteína diana no necesita ser purificado proporcionando los controles negativos apropiados de panning se han utilizado. Incluso objetivo variantes de proteínas que sólo están presentes en concentraciones muy bajas en el material biológico complejo se puede utilizar en la etapa de panning positivo. A través de la aplicación de esta tecnología, hemos adquirido anticuerpos contra proteínas variantes de TDP-43 que se encuentran de forma selectiva en el tejido cerebral ALS humana. Esperamos que este protocolo debe ser aplicable a la generación de reactivos que se unen selectivamente variantes de proteínas presentes en una amplia variedad de diferentes procesos biológicos y enfermedades.

En la Figura 1, el esquema muestra el proceso paneo negativo por el cual hemos eliminado fago de unión fuera de objetivo antígenos de nuestra biblioteca utilizando inmunotubos.
