Oxidatieve stress is geassocieerd met vele fysiologische en pathologische processen, alsmede xenobiotische metabolisme, wat leidt tot de oxidatie van biomacromoleculen, zoals DNA. Daarom efficiënte detectie van DNA oxidatie is belangrijk voor verschillende onderzoeksdisciplines, inclusief medicijnen en toxicologie. Een gemeenschappelijke biomarker oxidatief beschadigde DNA 8-oxo-7,8-dihydro-2&#39;-deoxyguanosine (8-oxo-dGuo; vaak verkeerd aangeduid als 8-hydroxy-2&#39;-deoxyguanosine (8-OH-dGuo of 8 oxo-dG)). Verschillende protocollen voor 8-oxo-dGuo meting door hogedruk vloeistofchromatografie met elektrochemische detectie (HPLC-ED) zijn beschreven. Deze werden echter voornamelijk toegepast op gezuiverde DNA behandeld met pro-oxidanten. Bovendien vanwege methodologische verschillen tussen laboratoria, voornamelijk vanwege het verschil in de analytische apparatuur, de toepassing van gepubliceerde werkwijzen voor detectie van 8-oxo-dGuo HPLC-ED vereist een zorgvuldige optimalisatie door elk laboratorium. EENuitgebreide protocol, beschrijft een dergelijk optimalisatieproces, ontbreekt. Hier, wordt een gedetailleerd protocol beschreven voor de detectie van 8-oxo-dGuo door HPLC-ED, in DNA van gekweekte cellen of dierlijke weefsels. Het illustreert hoe DNA monstervoorbereiding gemakkelijk en snel kan worden geoptimaliseerd om ongewenste DNA oxidatie die kunnen optreden tijdens monsterbereiding te minimaliseren. Dit protocol toont hoe 8-oxo-dGuo detecteren in gekweekte humane alveolaire adenocarcinoma cellen (bijv A549 cellen) behandeld met het oxidatiemiddel KBrO 3, en de milt van muizen die zijn blootgesteld aan de polycyclische aromatische koolwaterstoffen dibenzo (def, p) chryseen (DBC, voorheen bekend als dibenzo (a, l) pyreen, DalP). Kortom, dit werk illustreert hoe een HPLC-methode ED gemakkelijk kan worden geoptimaliseerd voor de detectie van 8-oxo-dGuo in biologische monsters.

dGuo werd waargenomen dat een retentietijd van 4,7 min, terwijl 8-oxo-dGuo had een retentietijd van ongeveer 6,4 min (figuur 2A en B) hebben.
