Développement de thérapies pour cibler les cellules disséminées nécessite une compréhension des mécanismes de la colonisation métastatique, la première étape critique dans le développement de la maladie péritonéale. Contrairement à la plupart des tumeurs solides, des métastases de cancer de l&#39;ovaire séreux de haut grade (HGSC) sont limités à la cavité péritonéale 1. Ainsi, les thérapies efficaces péritonéale pourraient contrôler ou d&#39;éradiquer HGSC. Actuellement, une approche thérapeutique norme est cytoréduction chirurgicale combinée avec la chimiothérapie 1-3. Malheureusement, la grande majorité des patients expérience et succomber à des complications de récurrence de la maladie. Ces statistiques lamentables montrent la nécessité d&#39;améliorer la compréhension de la colonisation métastatique, le processus par lequel les cellules cancéreuses à localiser, utiliser, et proliférer dans les tissus de l&#39;hôte pour former des métastases 2. L&#39;épiploon est un site début préféré de HGSC métastases 4-7. Contrairement à d&#39;autres adipeux péritonéale, le tissu adipeux épiploïque contient des structures inhabituelles immunitaires appelées taches laiteuses, qui contiennent B, T et les cellules NK et les macrophages, qui jouent des rôles clés dans Homeos péritonéalesTASIS 8,9. En plus de leurs fonctions physiologiques, nous avons constaté que des taches laiteuses jouent un rôle actif dans le cancer ovarien métastatique 4 colonisation. Dans les dosages de métastases expérimentales, SKOV3ip.1, Caov3, et HeyA8 (humain) et ID8 (murine; C57BL / 6) cellules de cancer ovarien rapidement à domicile points laiteux, ce qui suggère que les cellules se dirigent vers un facteur chimiotactique sécrétée. Fait intéressant, les cellules cancéreuses ne colonisent adipeux péritonéale manque taches laiteuses (c.-à-gonadiques et de la graisse de l&#39;utérus) 4. Un avantage particulier des approches décrites ici est l&#39;accent mis sur l&#39;architecture et la fonction des tissus, qui permet aux utilisateurs de tester des hypothèses dans entièrement intégré in vivo et ex vivo modèles de metastatic 4,10 de la colonisation. En comparant la localisation des cellules cancéreuses et la croissance dans les dépôts de graisse péritonéale qui soit contiennent ou manquent des taches laiteuses, les enquêteurs peuvent tester la contribution (s) relative des adipocytes et des cellules à l&#39;intérieur des taches laiteuses à l&#39;hébergement et de la croissance progressive des cellules du cancer de l&#39;ovaire dans les tissus physiologiquement pertinents.

in vivo and ex vivo approaches to study ovarian cancer metastatic colonization of milky spot structures in peritoneal adipose

Haute qualité de l&#39;ovaire séreux cancer (HGSC), la cause de métastases péritonéales répandues, continue d&#39;avoir un très mauvais pronostic; moins de 30% des femmes sont en vie 5 ans après le diagnostic. L&#39;épiploon est un site privilégié de la formation de métastases HGSC. Malgré l&#39;importance clinique de cette microenvironnement, la contribution du tissu adipeux épiploïque à la progression de cancer de l&#39;ovaire reste peu étudié. Omental adipeux est inhabituel en ce qu&#39;il contient en tant que structures connues taches laiteuses, qui sont constitués de B, T, et les cellules NK, des macrophages et des cellules progénitrices autour de nids denses de système vasculaire. Taches laiteuses jouent un rôle clé dans les fonctions physiologiques de l&#39;épiploon, qui sont nécessaires pour l&#39;homéostasie péritonéale. Nous avons montré que des taches laiteuses favorisent également le cancer de l&#39;ovaire métastatique de la colonisation adipeux péritonéale, une étape clé dans le développement de métastases péritonéales. Nous décrivons ici les approches que nous développés pour évaluer et quantifier les taches laiteuses en paritoneal adipeux et étudier leur contribution fonctionnelle à la cellule de cancer de l&#39;ovaire métastatique colonisation des tissus omentales fois in vivo et ex vivo. Ces approches sont plus généralisables à des modèles de souris et des lignées cellulaires, ce qui permet l&#39;étude de l&#39;ovaire de la formation de métastases de cancer localisation initiale des cellules de structures ponctuelles laiteux pour le développement de métastases peritoneales généralisées.
