Majoriteten av framsteg inom cancerterapi kräver tester i djurmodeller innan kliniska prövningar. Möjligheten att studera tumörbiologi in vivo, att dra nytta av djurmodeller, utgör ett viktigt verktyg för forskare som undersöker cancer patogenes, som gör det möjligt att bedöma olika behandlingsmetoder. Urinblåsecancer är en av de vanligaste cancerformerna i urogenitalområdet, med nästan 75.000 nya fall varje år i USA 1. Höga hastigheter av återfall kräver livslång uppföljning, vilket gör blåscancer en av de mest kostsamma cancer att behandla. Guldmyntfoten behandling för hög kvalitet NMIBC är trans-uretral resektion följt av intravesikal immunoterapi med BCG. Även om den exakta mekanismen för anti-tumöraktivitet av BCG står att helt klarlagd, är det accepterat att aktiveringen av ett cellförmedlat immunsvar anrikad på T-hjälpar- (Th) 1 och cytotoxisk T (Te) 1-celler är avgörande för framgången av terapin 2. Trots att BCG förblir behandlingen av valmöjligheten för NMIBC, en hög andel patienter svarar inte på behandlingen; dessutom kan den orsaka ett antal biverkningar: ca 70% av behandlade tumörer återkomma efter en tid och ~ 15% framsteg till muskeln-invasiv form avsjukdom. Andra biverkningar i samband med BCG behandling inkluderar dysuri, cystit och njurinfektion 3-6. Utvecklingen av nya terapeutiska strategier måste ta hänsyn till användningen av prekliniska modeller, efter inledande in vitro-bedömning; Detta är särskilt relevant i tumörer vars mikro kan väsentligt påverka deras utveckling och lyhördhet för behandling. Under det senaste årtiondet har vi riktat flera aspekter av immunmodulerande aktiviteten hos HP-NAP, ett protein som produceras av bakterien Helicobacter pylori, som ursprungligen identifierats som kan främja endotel vidhäftning av polymorfonukleära celler (PMN) 7. Strukturellt tillhör HP-NAP till DNA-skyddande protein under utsvultna förhållanden (DPS) familj 8 och består av 12 identiska subenheter arrangerade i en dodecameric skal. Vi har visat att HP-NAP ären Toll-liknande receptor (TLR) 2-agonist, med ett starkt immunmodulerande aktivitet är ansvarig för drivning av differentieringen av T-lymfocyter mot Th1-fenotypen, både in vitro och in vivo 9-10. I samband med denna verksamhet, är HP-NAP kan omdirigera Th2 immunsvar i mer fördelaktigt Th1 svar i en musmodell av allergisk astma 11. För att utvärdera Th1-beroende antitumörpotential HP-NAP, vi drog fördel av en musmodell av cancer i urinblåsan utvecklat flera år sedan av O&#39;Donnel och kollegor 12 för att utvärdera effekterna av BCG administrationen. Med detta protokoll, vi visar att HP-NAP har en stark antitumörpotential mot cancer i urinblåsan och att effekten av HP-NAP administration parallellt med signifikant ackumulering, inom tumör och regionala lymfkörtlar, både Th1 och TC1-lymfocyter som producerar interferon ( IFN) -γ 13 </sup&gt;. Tumörer som isolerats från HP-NAP-behandlade möss visade mer nekros och mindre vaskularisering än den obehandlade motsvarighet. Denna rapport ger en stepbystep protokoll, med uppgifter om beredningen av djuren, deras urinrörets kateterisering, den kemiska förbränningen krävs för att fästa celler till urinblåsan slemhinnan, och injektion av tumörcellerna. Vi beskriver även topisk administration av HP-NAP som kan anses som en prototyp av eventuella terapeutiska medel som utvecklats för behandling av cancer i urinblåsan. Det bevis som erhållits genom att jämföra tumörer isolerade från kontroll och HP-NAP-behandlade djur betonar inte bara det faktum att HP-NAP kan vara en bra kandidat för cancer i urinblåsan immunterapi, men också den allmänna effektiviteten hos den experimentella inrättas.

evaluation of the efficacy of the h. pylori protein hp-nap as a therapeutic tool for treatment of bladder cancer in an orthotopic murine model

Bladder cancer is one of the most common malignancies of the urogenital tract. Intravesical injection of Bacillus Calmette-Gu&#233;rin (BCG) is the gold standard treatment for the high-grade non-muscle invasive bladder cancer (NMIBC). However, since the treatment-related side effects are relevant, newer biological response modifiers with a better benefit/side effects ratio are needed.
The tumour microenvironment can influence both tumour development and therapy efficacy. In order to obtain a good model, it is desirable to implant tumour cells in the organ from which the cancer originates.
In this protocol, we describe a method for establishing a tumour in the bladder cavity of female mice and subsequent delivery of therapeutic agents; the latter are exemplified by our use of Helicobacter pylori neutrophil activating protein (HP-NAP). A preliminary chemical burn of the mucosa, followed by the injection of mouse urothelial carcinoma cell line MB49 via urethral catheterization, enables the cells to attach to the bladder mucosa. After a period, required to allow an initial proliferation of the cells, mice are treated with HP-NAP, administrated again via catheterization. The anti-tumour activity of HP-NAP is evaluated comparing the tumour volume, the extent of necrosis and the degree of vascularization between vehicle- and HP-NAP-treated animals.
