les dépôts amyloïdes de plaques, qui sont composés de bêta-amyloïde (Aß 1-42), sont un élément clé de la pathologie de la maladie d&#39;Alzheimer (MA). De nombreuses études ont montré que des niveaux élevés ou toxiques d&#39;oligomères Aß recombinant 1-42 susciter la mort neuronale, la dystrophie synaptique, la perte et la dysfonction; ainsi que l&#39;apprentissage et la mémoire déficits 1-4. Les régions du cerveau touchées comprennent l&#39;hippocampe, le cortex et des structures sous-corticales telles que le cerveau antérieur basal et 5,6 de l&#39;amygdale. À ce jour, il existe des modèles de souris transgéniques multiples qui tentent de simuler l&#39;Aß 1-42 pathologie de la MA. Malheureusement, à l&#39;exception de deux lignées transgéniques, APP23 et 5XFAD, ces souris ne répliquent intégralement la perte neuronale, un aspect clé de la MA. Même avec la perte neuronale observée dans APP23 et 5XFAD, l&#39;obser de mort neuronalecb était subtil, dépend de l&#39;âge, et isolée à quelques régions de sélectionner 7,8. La perfusion directe de oligomères Aß 1-42 dans le cerveau de souris de type sauvage fournit un excellent modèle in vivo, qui reproduit l&#39;aspect de la mort neuronale de amyloidopathy 1,9,10. Contrairement aux modèles de souris transgéniques communément utilisées, la Aß 1-42 oligomère modèle de perfusion est idéal pour l&#39;élévation aiguë Aß 1-42 niveaux de manière spatiale et temporelle. L&#39;avantage d&#39;utiliser les souris de type sauvage pour ce modèle évite les effets de compensation ou secondaires potentiels des mutations introduites dans les lignées de souris transgéniques. Des études antérieures ont montré que la perfusion des niveaux toxiques de Aß 1-42 dans l&#39;hippocampe provoque la mort des neurones au voisinage du point d&#39;injection pendant 1 semaine 1. La maladie d&#39;Alzheimer est une maladie neurodégénérative qui affecte le vieillissement de la population. Une caractéristique clé neuropathologiques de la maladie est la sur-production de bêta-amyloïde et le dépôt de plaques amyloïdes bêta dans les régions du cerveau des personnes souffrantes. Tout au long des années, les scientifiques ont généré de nombreux modèles de souris de la maladie d&#39;Alzheimer qui tentent de reproduire la pathologie bêta-amyloïde. Malheureusement, les modèles de souris seulement imitent de manière sélective les caractéristiques de la maladie. La mort neuronale, un effet important dans le cerveau des patients atteints de la maladie d&#39;Alzheimer, est visiblement défaut dans ces souris. Par conséquent, nous et d&#39;autres avons employé une méthode d&#39;infuser directement espèces oligomères solubles de bêta-amyloïde - formes de bêta-amyloïde qui ont été prouvés pour être plus toxique pour les neurones - stéréotaxique dans le cerveau. Dans ce rapport, nous utilisons mâles C57BL / 6J pour documenter cette technique chirurgicale de l&#39;augmentation des niveaux bêta-amyloïde dans une région du cerveau sélectionnez. Lecible de perfusion est le gyrus denté de l&#39;hippocampe, car cette structure du cerveau, ainsi que le cerveau antérieur basai qui est connectée par le circuit cholinergique, représente l&#39;un des domaines de dégénérescence dans la maladie. Les résultats de l&#39;élévation de bêta-amyloïde dans le gyrus denté par l&#39;intermédiaire d&#39;infusion stéréotaxique révèlent une augmentation de la perte de neurones dans le gyrus denté pendant 1 semaine, alors qu&#39;il ya une augmentation concomitante de la mort cellulaire et cholinergique perte de neurones dans la branche verticale de la bande diagonale de Broca du cerveau antérieur basal. Ces effets sont observés jusqu&#39;à 2 semaines. Nos données suggèrent que le modèle actuel de perfusion bêta-amyloïde fournit un modèle de souris alternative à aborder la mort des neurones spécifiques de la région en une base à court terme. L&#39;avantage de ce modèle est que les bêta-amyloïde peut être élevé d&#39;une manière spatiale et temporelle.
