Somatik hücreler, dört transkripsiyon faktörlerinin (Ekim-4, Sox-2, Klf-4 ve c-myc) ekspresyonunu zorlayarak uyarılmış pluripotent kök hücreler (iPSCs) içine yeniden olabilir, tipik olarak insan embriyonik kök hücreleri tarafından (hESC) olarak ifade . Nedeniyle hESC ile benzerliği, iPSCs hESC ile ilgili etik sorunlar kaçınarak, olası hastaya özgü rejeneratif tıp için önemli bir araç haline gelmiştir. Klinik uygulamalar için, uygun hücreler elde etmek için, transgen içermeyen iPSCs transgen reaktivasyonu, değiştirilmiş gen ekspresyonunu ve yanlış farklılaşmasını önlemek için söz konusu olacaktır. Ayrıca, yüksek ölçüde verimli ve pahalı olmayan bir yeniden programlama yöntemi, terapötik amaçlar için yeterli iPSCs elde etmek gereklidir.
