O protocolo aqui apresentado permite a purificação de vectores HCAdV com base em adenovírus humano tipo 5 com base em procedimentos descritos anteriormente 4,12. Para aplicações de terapia génica, é de grande importância para evitar efeitos colaterais citotóxicos e imunogénicas provocadas pela expressão de proteínas virais, a própria transgene ou por proteínas virais de entrada. Os vectores de adenovírus (Adv) são amplamente utilizados para introduzir ADN estranho numa ampla variedade de células para investigar o impacto da expressão do transgene 1,2. A versão mais avançada do AdV é representado por adenovírus vetores de alta capacidade (HCAdV) que faltam todas as sequências codificadoras virais 3,4 e oferecendo assim uma capacidade de acondicionamento até 35 kb, combinadas com baixa imunogenicidade e baixa toxicidade 5-8. Devido à sua capacidade de embalagem de alta permitem a entrega de transgenes grandes ou múltiplas utilizando uma dose única do vetor. Portanto, eles representam uma ferramenta valiosa para a comunidade de pesquisa. Em contraste com a primeira ou segunda geração AdV faltam os genes precoces E1 e / ou E3 que podem ser facilmente produzidos utilizando-se kits comerciais, vecTor construção do genoma do vírus e a produção de HCAdV é mais complexo. O sistema para a construção de genomas HCAdV baseia-se na pAdFTC plasmídeo que transporta um genoma HCAdV desprovido de todos os sequências codificadoras virais e o plasmídeo de transporte pHM5 9-12. Por conseguinte, um gene clonado de interesse possa ser libertado por PI- consecutivo Sce I e I-CEU I digere para a subsequente inserção dirigida para os mesmos sítios de restrição presentes no genoma HCAdV contido no plasmídeo pAdFTC. Em pAdFTC o local de inserção do transgene localizado entre a PI-Sce I e I-I Ceu sítios de clivagem é flanqueado por ADN de enchimento e as sequências não codificantes adenovirais necessárias para o empacotamento do genoma tais como repetições terminais 5 &#39;e 3&#39; invertidas (ITRs)em ambas as extremidades e o sinal de empacotamento a jusante do 5&#39;ITR. O DNA stuffer adicional fornece tamanho ideal do genoma HCAdV finais variando de 27 a 36 kb para garantir a embalagem eficiente durante a produção do vírus. Desde pAdFTC é um grande plasmídeo com até 45 kb (dependente do tamanho do transgene inserido) e o uso de endonucleases homing com locais de reconhecimento de ADN de comprimento comparativamente exibe forte ligação ao ADN, várias etapas de limpeza são necessárias durante a transferência do transgene a partir de pHM5 para pAdFTC. É recomendado um tratamento cuidadoso evitando forças de cisalhamento. As ITRs do genoma HCAdV são acompanhadas por Nem eu locais de reconhecimento de enzimas de restrição situados directamente a montante do 5&#39;ITR ea jusante da 3&#39;ITR 12. Portanto, HCAdV pode ser libertada por digestão Not I para a transfecção subsequente do genoma viral para a linha celular produtora HCAdV. Note-se que a utilização do enzima de restrição Not I para o relfacilidade do genoma viral a partir do plasmídeo pAdFTC implica que o transgene inserido é desprovido de sítios de reconhecimento de Not I de ADN. As células produtoras baseadas em células HEK293 (116 células) expressam de forma estável Cre recombinase. Para a amplificação do vírus 116 células são co-infectados com um vírus auxiliar (HV) fornecendo todos os genes necessários para a replicação AdV e embalagem de trans 3,4. O HV é um AdV de primeira geração com um sinal de empacotamento floxed que é removido durante a amplificação por vírus recombinase Cre expresso em células 116 4. Isto assegura que os genomas predominantemente HCAdV contendo um sinal de empacotamento está intacto encapsidado. Pré-amplificação do HCAdV é efectuada através da realização de passagens em série em passos de 116 células cultivadas em superfícies em pratos de cultura de tecidos. Depois de cada passagem partículas virais são libertadas a partir de células infectadas através da realização de três passos consecutivos de congelação-descongelação. Com cada passagem o aumento do número de células são infectadas com1/3 de lisado celular a partir da passagem anterior. Finalmente lisado proveniente do último passo de pré-amplificação é utilizado para infectar células produtoras cultivadas em suspensão num balão rotativo para a amplificação de grande escala. Os viriões são purificadas a partir das células em suspensão através da realização de uma ultracentrifugação em gradiente de densidade de cloreto de césio 4,12. Com este procedimento, as partículas vazias e partículas completamente montados são separados em duas bandas distintas. Para concentrar ainda mais o HCAdV partículas de um segundo passo de ultracentrifugação não gradual, é executada. Subsequentemente, a banda resultante contendo o HCAdV é recolhido e dialisado contra um tampão fisiológico. Preparações de vector finais são caracterizados no que diz respeito ao número de partículas virais infecciosas absolutos, partículas e os níveis de contaminação HV. High-capacity adenoviral vectors (HCAdV) devoid of all viral coding sequences represent one of the most advanced gene delivery vectors due to their high packaging capacity (up to 35 kb), low immunogenicity, and low toxicity. However, for many laboratories the use of HCAdV is hampered by the complicated procedure for vector genome construction and virus production. Here, a detailed protocol for efficient cloning and production of HCAdV based on the plasmid pAdFTC containing the HCAdV genome is described. The construction of HCAdV genomes is based on a cloning vector system utilizing homing endonucleases (I-CeuI and PI-SceI). Any gene of interest of up to 14 kb can be subcloned into the shuttle vector pHM5, which contains a multiple cloning site flanked by I-CeuI and PI-SceI. After I-CeuI and PI-SceI-mediated release of the transgene from the shuttle vector the transgene can be inserted into the HCAdV cloning vector pAdFTC. Because of the large size of the pAdFTC plasmid and the long recognition sites of the used enzymes associated with strong DNA binding, careful handling of the cloning fragments is needed. For virus production, the HCAdV genome is released by NotI digest and transfected into a HEK293 based producer cell line stably expressing Cre recombinase. To provide all adenoviral genes for adenovirus amplification, co-infection with a helper virus containing a packing signal flanked by loxP sites is required. Pre-amplification of the vector is performed in producer cells grown on surfaces and large-scale amplification of the vector is conducted in spinner flasks with producer cells grown in suspension. For virus purification, two ultracentrifugation steps based on cesium chloride gradients are performed followed by dialysis. Here tips, tricks, and shortcuts developed over the past years working with this HCAdV vector system are presented.

Cloning and Large-Scale Production of High-Capacity Adenoviral Vectors Based on the Human Adenovirus Type 5

A protocol for generation of high-capacity adenoviral vectors lacking all viral coding sequences is presented. Cloning of transgenes contained in the vector genome is based on homing endonucleases. Virus amplification in producer cells grown as adherent cells and in suspension relies on a helper virus providing viral genes in trans.
