Det finnes flere programmer av sulfidogenesis i miljøbioteknologi, en av de mest brukte programmene av metabolismen av sulfatreduserende bakterier i konsortier med fermen bakterier er i avløpsrensing. UASB reaktorer er blant de viktigste konstruerte tilnærminger til industrielt avløpsvann med høye sulfatkonsentrasjoner. I dette arbeidet vil vi presentere en protokoll for å få sulfidogenic slam fra marine sedimenter i en UASB reaktor. De kritiske trinnene i protokollen for å oppnå en sulfidogenic slam fra marine...

En av de viktigste bidrag til miljø bioteknologi var utformingen av bioreaktorer, hvor slammet som brukes (inoculum) var i stand til å utføre i henhold til sulfat reduserende betingelser. Sulfat reduksjon (SR) tillater rensing av avløpsvann strømmer som inneholder høye konsentrasjoner av sulfat i tillegg til den samtidige fjerning av COD, tungmetaller og organiske miljøgifter, et faktum som gjør SR en ønskelig egenskap av slammet 1. Noen eksempler på utslipp forurenset med sulfat kommer fra garveri, papir, farmasøytisk og kjemisk industri 1. Men de fleste av litteraturen viser til sulfidogenic slam når metanogenisk granulert slam har blitt tilpasset til sulfidogenesis 2. Denne tilpasningen er vanligvis oppnådd ved å manipulere COD / SO 4 2- forholdet i bioreaktoren og tilsetning av kjemikalier for å hemme metanogener i slammet 2,3. I tillegg til den lange tid som may krever dannelsen av sulfidogenic granuler, konkurransen mellom metanogener og sulfatreduserende midler og toleransen av slammet for høye konsentrasjoner av sulfid er noen av de viktigste problemene som kan oppstå dersom sulfidogenic slam som brukes i bioreaktoren er hentet fra tilpasning av overveiende metanogenisk slam til sulfatreduserende forhold. I dette arbeidet, beskriver vi fremgangsmåten for å få en overveiende sulfidogenic slam fra hydrotermale sedimenter (Punta Mita, Nayarit, Mexico) i en oppstrøms anaerob slamteppe reaktor (UASB), så vi evaluere sitt sulfatreduserende aktivitet over tid og gjennomføre et eksperiment for å evaluere dens anvendelse ved reduktiv dechlorination. Plasseringen av sedimentene ble valgt fordi det er blitt rapportert at i dette området er det dannelse av sulfider på grunn av den sulfatreduserende aktivitet som utvises av den mikrobielle samfunnet bebor det aktuelle sted 4. Det er several fordeler med å få dette sulfidogenic slam fra sedimenter i løpet tilpasse metanogenisk granulert slam til sulfidogenesis. Noen av disse fordeler er: (1) det er ikke nødvendig for å danne granuler for bioreaktoren å operere, (2) den slam tåler forholdsvis høye konsentrasjoner av sulfid i forhold til andre UASB som opererer med tilpasset metanogenisk slam, og (3) det er ingen konkurranse for underlaget med metanogener selv om acetat anvendes i en blanding av flyktige fettsyrer som inngår i kulturmediet for å fremme dannelsen av slam. Denne prosedyren ble fulgt for å fremme sulfidogenesis fordi marine sedimenter er et naturlig basseng av et bredt spekter av mikroorganismer som sulfatreduserende bakterier, fermen bakterier og dehalogenating bakterier bare for å nevne noen få 5,6. Den type konsortium utviklet fra marine sedimenter ved hjelp av denne protokollen kan utvise effektivitet i sulfat reduksjon, og derfor høye s<html> ulfate redusere aktivitet over tid og høyere toleranse for sulfid i konsentrasjoner høyere enn rapportert som giftig for metanogener og sulfatreduserende bakterier. På den annen side er det sannsynlig at dehalogenating evne er også vist i sedimentene ved å følge protokollen foreslått her, men det kan være avhengig av den opprinnelige mikrobielle samfunnet. Denne antakelsen er gjort basert på det faktum at reduktive dechlorination kan skje enten ved respirasjon eller cometabolism, begge forhold som kan bli fremmet i det marine mikrobielle samfunn 7. Dyrking av sedimentene for å oppnå slammet ble utført ved anvendelse av en blanding av acetat, propionat og butyrat som substrat, fordi disse flyktige fettsyrer blir brukt av flere stammer av sulfatreduserende bakterier. Disse syrene er også typen av karbonforbindelser som ofte finnes i marine sedimenter, ifølge flere rapporter i litteraturen med karbonholdig materiale i marine sedimenter 5,6. innhold &quot;&gt; Endelig er noen av de mest giftige forbindelser som finnes i grunnvann og andre vassdrag i verden er de klorerte løsningsmidler slik som trikloretylen (TCE) eller perkloretylen (PCE). Disse forbindelser er giftige, ikke bare for mennesket, men også for mikroorganismer, særlig TCE, som fortsatt betraktet som en prioritet forurensende av Environmental Protection Agency i USA 8. I dette arbeidet foreslo vi et eksperiment hvor sulfidogenic slammet blir testet på sin evne til å redusere TCE ved konsentrasjoner som er i spredningen for klorerte forbindelser biologisk nedbrytning i henhold metanogenisk forhold 9,10. Det er verdt å nevne at det meste av forskningen på biologisk nedbrytning av klorerte forbindelser har blitt gjennomført under metanogenisk forhold 9,10. Vi anser at forsøket med TCE foreslått i denne protokollen er en godt eksempel på de potensielle anvendelser av slammet. Formålet med dette eksperimentet var å evaluate toleransen av slammet til TCE og TCE virkning på sulfatreduserende aktivitet. Tatt i betraktning at det meste av forskningen på biologisk nedbrytning av klorerte forbindelser blir utført under metanogenisk forhold, foreslår denne protokollen dannelse av et slam kan brukes for samtidig å: (1) å fjerne sulfat, (2) fjerne COD og (3) fjerne klorerte forbindelser. Et ytterligere trinn kan være å evaluere slammet på den samtidige fjerning av TCE og tungmetaller (i tillegg til sulfat og COD), to forhold som ikke kan bli evaluert under metanogenisk betingelser.

The importance of microbial sulfate reduction relies on the various applications that it offers in environmental biotechnology. Engineered sulfate reduction is used in industrial wastewater treatment to remove large concentrations of sulfate along with the chemical oxygen demand (COD) and heavy metals. The most common approach to the process is with anaerobic bioreactors in which sulfidogenic sludge is obtained through adaptation of predominantly methanogenic granular sludge to sulfidogenesis. This process may take a long time and does not always eliminate the competition for substrate due to the presence of methanogens in the sludge. In this work, we propose a novel approach to obtain sulfidogenic sludge in which hydrothermal vents sediments are the original source of microorganisms. The microbial community developed in the presence of sulfate and volatile fatty acids is wide enough to sustain sulfate reduction over a long period of time without exhibiting inhibition due to sulfide. 
This protocol describes the procedure to generate the sludge from the sediments in an upflow anaerobic sludge blanket (UASB) type of reactor. Furthermore, the protocol presents the procedure to demonstrate the capability of the sludge to remove by reductive dechlorination a model of a highly toxic organic pollutant such as trichloroethylene (TCE). The protocol is divided in three stages: (1) the formation of the sludge and the determination of its sulfate reducing activity in the UASB, (2) the experiment to remove the TCE by the sludge, and (3) the identification of microorganisms in the sludge after the TCE reduction. Although in this case the sediments were taken from a site located in Mexico, the generation of a sulfidogenic sludge by using this procedure may work if a different source of sediments is taken since marine sediments are a natural pool of microorganisms that may be enriched in sulfate reducing bacteria.
