Først introdusert i 2006, benytter DNA origami selv montering natur DNA-oligonukleotider å produsere utformbare og svært bestilte nanostrukturer. 1 En myriade av strukturer har blitt rapportert, alt fra smilefjes til låst 3-dimensjonale bokser. 2 DNA origami kan funksjon med ulike biomolekyler og nanostrukturer, noe som gir opphav til forskningssøknader i nanoelektronikk, medisin og quantum computing. 3 Men analysen og mange fremtidige programmer er ikke bare avhengig av strukturelle design, men også på vedheft av DNA origami nanostrukturer til overflater. Metodene som beskrives i dette manuskriptet gjelder for fremstillingen av DNA-origami prøver på to typer substrater: glimmer og funksjonaliseres silisiumoksid. Glimmer er substratet grepet av DNA origami studier fordi det er atomisk flat, med en laghøyde på 0,37 nm ± 0,02 nm. 4 er det også easily rengjort, noe som gjør atomic force mikroskopi (AFM) studier prøveopparbeidelse og grei. Silicon passiv med et tynt innfødt oksid har attraktive elektroniske egenskaper, inkludert kompatibilitet med tidligere gratis Metal Oxide Semiconductor (CMOS) behandling for å lage input / output strukturer og topografiske egenskaper. Silisiumskiver som er lagret i luft er passivert med enten en tykk termisk oksyd eller tynn innfødt oksid film som er relativt skittent, med en høy partikkeltelling. På magnesium ionekonsentrasjoner above 150 mM, god dekning (opp til 4 / um 2) med rektangulært DNA origami kan oppnås på oksygenplasma behandles silisiumsubstrater; Dette kan imidlertid konsentrasjonen og dekning endres avhengig av størrelsen og utformingen av nanostrukturer som brukes. 10 En alternativ protokoll for tuning overflaten kostnad er å feste en kationiske selv-montert monolayer av 3-aminopropyltrietoksysilan (APTES) (figur 1B) til oksydet. The designed nature and controlled, one-pot synthesis of DNA origami provides exciting opportunities in many fields, particularly nanoelectronics. Many of these applications require interaction with and adhesion of DNA nanostructures to a substrate. Due to its atomically flat and easily cleaned nature, mica has been the substrate of choice for DNA origami experiments. However, the practical applications of mica are relatively limited compared to those of semiconductor substrates. For this reason, a straightforward, stable, and repeatable process for DNA origami adhesion on derivatized silicon oxide is presented here. To promote the adhesion of DNA nanostructures to silicon oxide surface, a self-assembled monolayer of 3-aminopropyltriethoxysilane (APTES) is deposited from an aqueous solution that is compatible with many photoresists. The substrate must be cleaned of all organic and metal contaminants using Radio Corporation of America (RCA) cleaning processes and the native oxide layer must be etched to ensure a flat, functionalizable surface. Cleanrooms are equipped with facilities for silicon cleaning, however many components of DNA origami buffers and solutions are often not allowed in them due to contamination concerns. This manuscript describes the set-up and protocol for in-lab, small-scale silicon cleaning for researchers who do not have access to a cleanroom or would like to incorporate processes that could cause contamination of a cleanroom CMOS clean bench. Additionally, variables for regulating coverage are discussed and how to recognize and avoid common sample preparation problems is described.
