El campo de la investigación nanotoxicología pretende caracterizar el efecto biológico de las nanopartículas. Los estudios toxicológicos sobre la base de las investigaciones in vivo siguen siendo los métodos más precisos. Sin embargo, su uso está limitado por sus requisitos de coste, tiempo y trabajo 1. Como los ensayos de alternativas, in vitro se han utilizado debido a su simplicidad y la posibilidad de desarrollar de alto rendimiento en plataformas de pruebas in vitro 2 - así se amplía el número de condiciones ensayadas. La mayoría de los estudios nanotoxicological se realizan usando ensayos in vitro con líneas celulares inmortalizadas. Sin embargo, hay preocupaciones con respecto a la extrapolación de estos resultados experimentales in vivo a los efectos toxicológicos 3. kupffer cell isolation for nanoparticle toxicity testing

La gran mayoría de los estudios in vitro nanotoxicological han utilizado líneas celulares inmortalizadas por su practicidad. Sin embargo, los resultados de las pruebas de toxicidad de nanopartículas en líneas celulares inmortalizadas o células primarias han mostrado discrepancias, destacando la necesidad de extender el uso de células primarias de ensayos in vitro. Este protocolo describe el aislamiento de los macrófagos del hígado del ratón, las células de Kupffer con nombre, y su uso para estudiar la toxicidad de nanopartículas. Las células de Kupffer son la población de macrófagos más abundante en el cuerpo y constituyen parte del sistema retículo-endotelial (RES), responsable de la captura de las nanopartículas de circulación. El método de aislamiento de células Kupffer informó aquí se basa en un método de perfusión de 2 pasos, seguido de purificación en gradiente de densidad. El método, basado en la digestión de colagenasa y centrifugación de densidad, es una adaptación del protocolo original desarrollada por Smedsrød et al. diseñado para la rata isolatio células hepáticasn y proporciona un alto rendimiento (hasta 14 x 10 6 células por ratón) y alta pureza (&gt; 95%) de las células de Kupffer.
