The syngeneic rat mesothelioma II-45 cell line was a kind gift from A/Prof. Emanuela Felley-Bosco, Zurich University.

Maligne mesothelioom (MM) is een agressieve maligniteit die voortvloeit uit getransformeerde cellen in het membraan (mesothelium) die lijnen de longen en de buikholte, hart en inwendige geslachtsorganen, en is de meest voorkomende primaire tumor van de long holte of pleura 1,2 . Blootstelling aan asbestvezels is goed voor 80% van alle MM, en terwijl het verbod op het gebruik van asbest werden decennia geleden geïntroduceerd in de meeste westerse landen, is het wijdverbreide gebruik in de gemeenschap een dodelijke erfenis. De Wereld Gezondheids Organisatie schat dat 107.000 mensen wereldwijd sterven elk jaar van asbest gerelateerde ziekten, met sterftecijfers blijft toenemen. Een nieuwe niet-beroepsmatige incidentie wave is ook in opkomst en er is weinig begrip voor wanneer en op welk niveau dit hoogtepunt zal bereiken 3. De meerderheid van de mensen met MM te laat gediagnosticeerd als systemische chemotherapie is een van de enige haalbare opties 4. De meest effective chemotherapie en de huidige &#39;standaard van zorg&#39; (samen pemetrexed met cisplatine 5) werd meer dan 10 jaar geleden geïdentificeerd. Maar het falen van deze behandeling is onvermijdelijk en zijn er geen bewezen tweede regel opties, waardoor patiënten met een grimmige prognose en mediane overleving van slechts 12 maanden 2. Daarom is er een dringende onvervulde behoefte aan effectievere behandelingen. Ondanks het onderzoek van een aantal nieuwe therapieën in klinische studies geen resulteert in veranderingen in de praktijk. Dit is deels te wijten aan de lage (5%) overdracht van pre-klinische resultaten, doorgaans bij xenograft muismodellen om de klinische instelling 6-8. Dergelijke modellen niet getrouw herhalen complexe aspecten van de tumor micro-omgeving voorkomen in niet-fysiologische locaties, vaak met afwezigheid van een functionerend immuunsysteem 9. Syngene orthotope modellen te maken een aanzienlijk realistischer tumor milieu dan de commonly gebruikt subcutane xenograft modellen als de tumoren zich in de juiste fysiologische locatie met een intact immuunsysteem 10,11. De grotere omvang van de rat verhoogt het gebruik als knaagdier ziektemodel, vooral in drug studies waarbij seriële bloedafnames moeten behandelingsrespons 12 en toxiciteit te beoordelen. Bovendien, in modellen waarin toezicht ziekteprogressie moeilijk vanwege de locatie van de tumoren (bijvoorbeeld in de pleurale holte), het vermogen tot ziekteprogressie controleren middels factoren die in het bloed is zeer aantrekkelijk. Het genereren van een syngene orthotope model borstvliesmesothelioma gebruik immuuncompetente ratten wordt beschreven. Bovendien wordt een eenvoudige en relatief niet-invasieve werkwijze voor het bewaken van pleurale ziekteprogressie door meting van circulerende immuuncellen ook beschreven.

De enorme toename van belangstelling immuun gebaseerde behandelingen voor kanker zoals vaccins en immune controlepunt remmers en beter begrip van de rol van de tumor micro-omgeving in behandelingsrespons collectief wijzen op de noodzaak van immuuncompetente orthotope modellen voor preklinische tests van deze nieuwe therapieën. Dit artikel laat zien hoe u een orthotope immuun-competente rat model van pleura maligne mesothelioom vast te stellen. Monitoring progressie van de ziekte in orthotopische modellen is beschaamd door de interne locatie van de tumoren. Om longitudinaal bewaken ziekteprogressie en zijn effect op circulerende immuuncellen in deze en andere rat modellen van kanker, een enkele buis flowcytometrie test die slechts 25 gl volbloed wordt beschreven. Dit verschaft kwantificatie van zeven immune parameters: totale lymfocyten, monocyten en neutrofielen, en T-cel subsets CD4 en CD8, B-cellen en natuurlijke killer cellen. Verschillende subsets van deze parameters zijn nuttig in verschillende omstandigheden en modellen, met de neutrofielen naar lymfocyt verhouding met de grootste hulpprogramma voor het monitoren van progressie van de ziekte in de mesothelioom model. Analyseren circulerende immuun cel niveaus met behulp van deze enkele buis methode kan ook helpen bij het toezicht op de respons op het immuunsysteem gebaseerde behandelingen en inzicht in de onderliggende mechanismen die leiden tot succes of falen van de behandeling.

De werkwijze die in dit document voor het genereren van een orthotopic model borstvliesmesothelioma gebruik II-45 cellen resulteerde bij dieren bezwijken mesothelioom in een reproduceerbare en snelle tijdsbestek, zonder ratten sterven als gevolg van de implantatie methode.
