Ondartet mesothelioma (MM) er en aggressiv malignitet som oppstår fra transformerte celler i membranen (mesothelium) som linjer i lungen og bukhulen, hjerte og indre kjønnsorganer, og er den mest vanlige primære svulst i tette hulrom eller pleura 1,2 . Eksponering for asbest fiber står for 80% av all MM, og mens forbud mot asbest bruk ble introdusert tiår siden i de fleste vestlige land, har utbredt bruk i samfunnet igjen en dødelig arv. Verdens helseorganisasjon har anslått at 107.000 mennesker over hele verden dør hvert år av asbestrelaterte sykdommer, med dødeligheten fortsetter å øke. En ny ikke-yrkes forekomst bølgen er også voksende, og det er liten forståelse for når og på hvilket nivå dette vil peak tre. Flertallet av mennesker med MM er diagnostisert sent når systemisk kjemoterapi representerer en av de eneste levedyktige alternativer 4. De fleste effective kjemoterapi og nåværende &quot;standard of care&quot; (pemetrexed sammen med cisplatin 5) ble identifisert over 10 år siden. Men svikt i denne behandlingen er uunngåelig, og det er ikke påvist andre linjen, forlater pasienter med dystre prognose og median overlevelse på bare 12 måneder 2. Derfor er det stort udekket behov for mer effektive behandlinger. Til tross for undersøkelse av en rekke nye behandlingsformer i kliniske studier har ingen ført til endringer i praksis. Dette skyldes delvis den lavt (5%) overføring av pre-kliniske resultater, vanligvis utført i xenograft musemodeller, for å klinisk setting 6-8. Slike modeller har ikke trofast repetere de komplekse deler av tumor mikromiljøet som forekommer i ikke-fysiologiske steder, ofte i fravær av et fungerende immunsystem 9. Den enorme utbruddet av interesse i immunbaserte behandlinger for kreft som vaksiner og immunsjekkpunkt hemmere, og økt forståelse av rollen av svulsten mikromiljøet i behandlingsrespons, kollektivt peke på behovet for immun-kompetente ortotopiske modeller for pre-klinisk testing av disse nye terapier. Dette papiret demonstrerer hvordan å etablere en orthotopic immunkompetente rotte modell av pleural ondartet mesothelioma. Overvåking sykdomsprogresjon i ortotopiske modeller er forvirret av den interne plasseringen av svulster.
