The use of polymeric micelles for drug delivery continues to expand due to their versatility and ability to deliver hydrophobic drugs for various disease states. Therefore, the techniques needed to prepare and characterize these formulations prior to use in cell culture or animals is a critical first step to determine the best pairing between the drug and the polymer. Amfifiele blokcopolymeren met herhalende structuur bestaande uit hydrofiele en hydrofobe domeinen spontaan zelfassemblage driedimensionale macromoleculaire samenstellingen zogenaamde polymere micellen te vormen. Deze structuren hebben een binnenste hydrofobe kern omgeven door een hydrofiele schil. De hydrofobe kern heeft de mogelijkheid om hydrofobe geneesmiddelen nemen door fysieke opsluiting tot hydrofobe interacties of door chemische conjugatie aan het polymeerskelet. 1 vele voordelen bestaan ​​om het gebruik van deze blokcopolymeren te micellen voor geneesmiddelafgifte vormen. Deze omvatten incorporatie van slecht oplosbare geneesmiddelen te verbeteren farmacokinetiek van de gebruikte geneesmiddelen en de biologische verenigbaarheid en / of biologische afbreekbaarheid van de polymeren maakt ze veilig alternatief voor conventionele oplosbaarmakende. 2 Een ander voordeel van het gebruik van polymere micellen hun colloïdale deeltjesgrootte tussen 15- 150 nm 3, waardoor ze aantrekkelijk voor parenteral levering. De omvang van deze micellen varieerde van 20 nm tot 85 nm. Het voordeel van PEG gebaseerde polymeren is hun biocompatibiliteit en afhankelijk van het tweede blok kan ook biologisch afbreekbaar zijn. Onlangs nieuwe geneesmiddelafgiftesystemen gebaseerd op polyethyleneglycol- blok -polylactic acid (PEG b -PLA) polymere micellen zijn ontwikkeld voor het gelijktijdig leveren van meerdere geneesmiddelen tegen kanker. De PEG-b- PLA micellen zijn zowel biocompatibel en biologisch afbreekbaar. Deze multi-geneesmiddel beladen micellen hebben getoond alsynergistic remming van verschillende kankers modellen in vitro en in vivo 2,5,6 en passen in het huidige paradigma van gebruik van meerdere geneesmiddelen bij chemotherapie resistentie of omlaagbrengen toxiciteit. Daarom is er veel belangstelling voor het bereiden en karakteriseren deze micellaire geneesmiddelafgiftesystemen voor gebruik bij kanker en andere ziekten. In het werk dat we hieronder geschetste stap-voor-stap proces waarbij dergelijke micellen kunnen worden bereid en gekarakteriseerd voor evaluatie daarvan bij ziektetoestanden plaats. Voor de toepassing van dit werk twee slecht oplosbare anti-kankermiddelen, docetaxel (DTX) en everolimus (EVR) zijn gekozen. Zowel DTX en EVR zijn slecht in water oplosbare verbindingen met intrinsieke oplosbaarheid in water bij 1,9 en 9,6 ug / ml. 7,8 Twee PEG-b -PLA polymeren met verschillende molecuulgewichten werden gebruikt in dit protocol als de bouwstenen voor de geformuleerde polymere micellen,deze polymeren zijn PEG 2000 - b -PLA 1800 (3800 Da) en PEG 4000 - b -PLA 2200 (6200 Da). PEG-b -PLA micellen kan dus zorgen voor een uniek platform als Nanodrager voor DTX en EVR afzonderlijk en in combinatie. Amfifiele blok-copolymeren als polyethyleneglycol- blok -polylactic zuur (PEG-b- PLA) kunnen zichzelf assembleren in micellen boven hun kritische micellaire concentratie vormen hydrofobe kernen omringd door hydrofiele schelpen in waterige omgevingen.
