De cellen die voor dfTAT afgifte moet niet al confluent zijn (&gt; 90% confluentie) omdat dit kan hebben op de levering efficiency. De cellen moeten ook gezond zijn: dode cellen in de cultuur kan apoptotische fragmenten waarmee dfTAT kan communiceren (bijvoorbeeld DNA van afgebroken kernen) vrij te geven. De afgifte van proteïnen, peptiden of cel-doorlaatbare kleine moleculen in levende cellen vaak gewenst in veel biologische of biotechnologische toepassingen (cellulaire beeldvorming, functionele testen, cellulaire herprogrammering, enz.) 1-4. Vele benaderingen levering reeds gemeld, waaronder micro-injectie, elektroporatie, of het gebruik van carrier middelen (bv., Celpenetrerende peptiden zoals TAT, lipiden) 5-7. Elke techniek heeft meestal specifieke voor- en nadelen die deze benaderingen voldoende voor sommige toepassingen, maar niet voor anderen kunnen maken. Interacties tussen dfTAT en MOI niet nodig zolang beide entiteiten verkeer tezamen in de endocytische route. Als dfTAT een bepaalde drempel binnen het lumen van endocytische organellen bereikt, zijn endosomale lekkage activiteit (de moleculaire details uitdrukt blijven volledig gekarakteriseerde te zijn). De inhoud van het lumen van lekkende organellen, en dus de MOI, wordt dan losgelaten in de cellen. Deze benadering is daarom erg handig omdat geen conjugatie of formulering stelsels met MOI vereist. Omdat bovendien dfTAT is niet direct modificeren van een MOI moet ook niet storen MOI functie eenmaal intracellulaire afgifte wordt bereikt. Ook de concentratie vandfTAT gebruikt levering is onafhankelijk van die van de MOI in de media. Zo kan dfTAT concentratie constant tussen verschillende experimenten worden gehouden reproduceerbare efficiëntie in endosomale lekkage te garanderen. Daarentegen kunnen concentratie MOI in media geleidelijk worden veranderd om gewenste niveau bereiken van MOI geleverd cytosol. De hoge endosomale lekkage efficiency bereikt met dfTAT opmerkelijk onschadelijk zijn voor veel van de tot op heden geteste cellen. Dit is verrassend omdat endosomale organellen belangrijke component van de cellen en men zou verwachten dat de dramatische lekkage veroorzaakt door dfTAT gepaard zou gaan met schadelijke cellulaire responsen. Toch behandelde cellen woekeren in hetzelfde tempo als onbehandelde cellen en geen significante veranderingen in hun transcriptoomanalyse niet weergegeven. Bovendien kan de levering worden herhaald binnen enkele minuten met reproduceerbare levering efficiëntie, wat aangeeft dat de cellen kan ofwel tolereren of herstellen van het leveringsproces zonder verlieshun capaciteit voor endocytose of endosomale lekkage. Subtiele cellulaire reacties kunnen plaatsvinden tijdens dfTAT levering en de moleculaire details van wat deze reacties zou kunnen nog worden onderzocht. Maar door het combineren hoge efficiëntie, gemak protocollen, en gebrek aan toxiciteit, dient deze levering benadering blijken onmiddellijk bruikbaar in vele cel-gebaseerde applicaties. De hierin gepresenteerde protocollen zijn gericht op het maken van deze technologie toegankelijk voor de onderzoeksgemeenschap.

Macromoleculaire levering strategieën gebruiken meestal de endocytische route als een route van cellulaire binnenkomst.
