Als deeltjes bindt, de resonantie golflengte (λ) van de torus toeneemt. Wanneer een deeltje unbinds de resonantiegolflengte dienovereenkomstig afneemt (een naar beneden event). Single molecule detectie experimenten ingezet om de hoeveelheid analyt die in biosensoren voor vroege detectie van ziekten en het onderzoek van de fundamentele eigenschappen van moleculen 3. Dergelijke experimenten worden typisch uitgevoerd met behulp labels echter etiketten niet altijd mogelijk verkrijgen voor een bepaald eiwit, verhogen de kosten, kan verstoren de gebeurtenissen bestudeerd en kan onhandig zijn, met name voor real time-eigen experimenten of point-of- care diagnostiek. De huidige gouden standaard voor ongelabelde biosensoren is oppervlakteplasmonresonantie 4, maar de commerciële oppervlakte plasmon resonance systemen hebben typisch een typisch onderste detectielimiet in de orde van nM. Onlangs hebben optische resonatoren ontpopt als een veelbelovende technologie voor het label-free single molecule BioDetection 5. Optische resonatoren werk gebaseerd op de lange termijn (ns) opsluiting van licht 6,7. Licht is evanescentlygekoppeld in deze apparaten meestal via een optische vezel. Wanneer de golflengte van het licht dat door de vezel komt overeen met de resonantiegolflengte van de resonator, licht efficiënt paren aan de resonator. Deze gekoppelde licht totaal intern reflecteert binnen de holte resonator genereren van een verdwijnende veld in de nabijheid van de omtrek van de resonator. Aangezien deeltjes komen het verdwijnende veld en binden aan de resonator, de resonantiegolflengte van de resonator verandert evenredig met het volume van het deeltje 8. Op het gebied van detectie vermogen, hebben microsferen resonatoren eerder is gebruikt om enkele influenza A-virus detecteren deeltjes (100 nm) 9,10. Recent zijn plasmonically versterkte microbolletjes optische resonatoren gebruikt om één runderserum albumine te detecteren moleculen 11 en 8-mer oligonucleotiden 12, maar deze benadering beperkt het deeltje capture gebied 0,3 um per 2 deondeugd. Groter gebied vast te leggen biosensoren zijn ideaal voor het maximaliseren van de kans op een deeltje detectie. Huidige-oplossing op basis-label vrij biosensing technologieën met grote (&gt; 100 micrometer 2) capture gebieden zijn beperkt tot het opsporen van polystyreen deeltjes ≥ 25 nm. We hebben een ongelabelde biosensoren gebaseerd op optische resonator techniek bekend als frequentievergrendeling Optical Whispering Evanescent Resonator (bloem) 13 (figuur 1) die in staat is tijdsopgeloste detectie van individuele moleculen in oplossing ontwikkeld. FLOWER maakt gebruik van de lange levensduur van foton microtoroıde optische resonatoren gecombineerd met een frequentie vergrendeling feedback control, evenwichtig detectie, en computationele filtering om kleine deeltjes te detecteren enkel eiwit moleculen. Het gebruik van frequentievergrendeling kan het systeem altijd volgen van de verschuiving van de resonantie microtoroıde als deeltjes binden, zonder te vegen of scan de laser golflengte bovengrote reeksen. De principes van de bloem kunnen worden gebruikt om de detectiemogelijkheden van andere technieken zoals plasmon enhancement verbeteren. Hierna worden de procedures voor het uitvoeren BLOEM beschreven.
