Ända sedan dess utveckling i mitten av 1990-talet SOD1-G93A transgen musmodell har varit den mest använda djurmodell av amyotrofisk lateralskleros (ALS) 1. Denna transgen musmodell genetiskt för att överuttrycka en mutant form av den humana Cu / Zn-superoxiddismutas 1 (SOD1) -genen härbärgerar ALS-associerade glycin till alaninmutation vid aminosyra 93 (G93A). Den G93A mutationen är en av många mutationer i SOD1-genen som tillsammans smink ungefär en femtedel av familjär ALS-fallen 2. Den SOD1-G93A modellen har blivit en arbetshäst i ALS läkemedelsutveckling forskning eftersom utöver dess uppenbara genetisk koppling till ALS, rekapitulerar många av de patologiska egenskaperna hos ALS hos människa såsom motorneuron förlust, progressiv muskelsvaghet och atrofi, med eventuell förlamning och död 3. Men som ofta är fallet med transgena djurmodeller, det finns en inneboende biologisk variabilitet associerad med SOD1-G93A möss. Scott et al., har fastställt att kohorter av minst 24 kull matchade könsbalanserade möss behövs läkemedelseffektstudiedesign för att övervinna det buller som skapats av den biologiska variabilitet 4. Det stora antalet djur som krävs i dessa studier i kombination med den aggressiva sjukdomsutveckling och behovet av daglig övervakning förbjuder användning av komplicerade och tidskrävande och potentiellt stressande metoder för att mäta neuromuskulär styrka och funktion (t.ex. elektromyografi, greppstyrka, rotorod , etc). I stället understryker det behovet av ett in vivo screening verktyg som på ett tillförlitligt sätt bedömer den neuromuskulära funktionen hos SOD1-G93A möss på ett snabbt sätt. Vid ALS TDI har ett protokoll som utvecklats som gör det möjligt för en tillförlitlig bedömning av den neuromuskulära funktionen hos SOD1-G93A möss på mindre än 30 sekunder per mus i genomsnitt. Detta protokoll encompaSSES daglig bedömning av bakdelen funktion med ett enkelt neurologisk poängsystem (NeuroScore) som beskrivs i denna rapport i press och video. NeuroScore fokuserar på bakdelen funktion eftersom bakdelen underskott är den tidigaste rapporterade neurologiska sjukdomstecken i SOD1-G93A möss 5,6. Vidare protokoll som utvecklats av ALS TDI ger en objektiv bedömning av uppkomsten av pares (lätt eller partiell förlamning), progression och svårighetsgraden av förlamning och det är tillräckligt känslig för att identifiera läkemedels inducerade förändringar i sjukdomsförloppet.

The SOD1-G93A transgenic mouse is the most widely used animal model of amyotrophic lateral sclerosis (ALS). At ALS TDI we developed a phenotypic screening protocol, demonstrated in video herein, which reliably assesses the neuromuscular function of SOD1-G93A mice in a quick manner. This protocol encompasses a simple neurological scoring system (NeuroScore) designed to assess hindlimb function. NeuroScore is focused on hindlimb function because hindlimb deficits are the earliest reported neurological sign of disease in SOD1-G93A mice. The protocol developed by ALS TDI provides an unbiased assessment of onset of paresis (slight or partial paralysis), progression and severity of paralysis and it is sensitive enough to identify drug-induced changes in disease progression. In this report, the combination of a detailed manuscript with video minimizes scoring ambiguities and inter-experimenter variability thus allowing for the protocol to be adopted by other laboratories and enabling comparisons between studies taking place at different settings. We believe that this video protocol can serve as an excellent training tool for present and future ALS researchers.
