LVs bieden belangrijke voordelen boven andere virale systemen voor de genetische modificatie van volwassen NSC 16,18. De muizen ventriculaire-subventriculaire zone (V-SVZ), in de wanden van de laterale ventrikel tegenover het striatum, is een zeer actief germinale gebied waarin een continu proces van voorlopercellen celdeling en differentiatie leidt tot de aanhoudende productie van bulbus olfactorius (OB ) interneuronen en corpus callosum oligodendrocyten 1. De levenslange genereren van deze cellen blijkt te worden door de aanwezigheid in deze regio van neurale stamcellen (NSC&#39;s; ook wel B1 cellen), waarbij de astrocytaire antigeen glia fibrillair zuur eiwit (GFAP) tot expressie en stamcel markers zoals nestine, Id1 en Sox2 2. GFAP expressie B1 cellen genereren transit versterken voorlopercellen (TAP) cellen (C-cellen), die tot expressie transcriptiefactoren Dlx2 (distale-less homeobox 2) en Ascl1 (zoogdieren Achaete-Schute homoloog 1) en verdeel snel een paar keer voordat ze aanleiding geven te migreren neuroblasts (A-cellen) of oligodendroblasts 3. Nieuw-gegenereerde ProlifERATIVE neuroblasts migreren naar voren, de vorming van de rostrale migratiestroom (RMS) aan de OB, waar ze te integreren in de granulaire en glomerulaire lagen zo gedifferentieerd remmende interneuronen. Migreren jonge oligodendroblasts verplaatsen naar de CC, waar ze worden onvolwassen NG2-positieve cellen die nog steeds lokaal splitsen of te differentiëren tot rijpe myeliniserende oligodendrocyten 1,4. B1 cellen, die afkomstig zijn van foetale radiale gliale cellen behouden langwerpig en gepolariseerde morfologie van hun voorgangers en vertonen een zeer gespecialiseerde relatie met hun niche. Omspant tussen de ependyma welke lijnen de ventrikel en het netwerk van bloedvaten die de V-SVZ niche irrigeren. De kleine apicale proces van B1 cellen intercalaten onder multiciliated ependymocytes en eindigt in een enkele niet-beweeglijke primaire cilium, terwijl hun basale proces strekt zich lange afstanden naar de vlakke vasculaire plexus dat deze niche einde bevloeit in de b benaderenasal lamina van de plexus haarvaten 2,5-8. De meest betrouwbare manier om B1-NSCs onderscheiden van niet-neurogene astrocyten, die ook GFAP +, in het intacte V-SVZ niche in gehele-mount preparaten van de ventrikel zijwand en de analyse van 3-D confocale microscopie na immunokleuring voor GFAP de dunne B1-NSC apicale proces, β-catenine label celmembranen af te bakenen, en ofwel γ-tubuline als een marker van cilial basale organen of geacetyleerde α-tubuline om het etiket van de omvang van elk cilium 5,8. Opmerkingen van deze hele mounts van de ventriculaire oppervlak hebben aangegeven dat B1 en ependymale cellen zijn opgesteld in &quot;vuurraderen&quot; 5, waarbij de uniciliated apicale processen van één of meer GFAP + B1 cellen worden omringd door een rozet van multiciliated ependymale cellen. De karakteristieke morfologie van B1 cellen correleert met experimenteel bewijs indicating die de bloedvaten / endotheelcellen en ventriculaire cerebrospinale vloeistof (CSF), vormen geregeld bronnen van oplosbare signalen die op NSC 2,6,9-11. Bij de ventriculaire oppervlak, homotypische en heterotypische apico-laterale interacties van ependymale en B1 cellen omvatten krappe kruispunten en adherens kruispunten 5,12. Bovendien adhesiemoleculen betrokken bij de junctionele complexen tussen B1 en ependymale cellen, zoals N-cadherine en V-CAM, is aangetoond dat niet alleen de zeer georganiseerde positionering van B1 regelen de V-SVZ niche, maar ook hun rust 12 , 13. De ependymale-B1 cel monolaag lijkt te werken als een diffusiebarrière waardoor de afgestelde stroom van water en kleine moleculen uit de CSF, maar beperkt de doorgang van grote intercellulaire eiwitten 10,11. Experimenteel bewijs geeft aan dat de unieke positie B1 cel apicale cilium een rol kunnen spelen als een sensor van signalering Polypeptiden aanwezig in de CSF 2,5-7. Ependymale cellen zijn op zich een bron van oplosbare en membraangebonden signalen met een rol in de regulatie van NSC gedrag 14,15. Traceerbaar nucleosiden, zoals broom- deoxyuridine (BrdU) of retrovirussen zijn op grote schaal gebruikt om progenitorcellen, waaronder NSCs label in vivo. Deze werkwijzen zijn niet optimaal voor de lange termijn gebeurt tracing omdat BrdU signalen verdund door herhaalde celdelingen en retrovirussen lijken preferentieel richten tijdelijk amplificeren van cellen als gevolg van de eis van celproliferatie voor transductie 16,17. Om NSC fysiologie in vivo, met inbegrip van interacties met niche componenten te onderzoeken, is het cruciaal om een methode om te labelen en te traceren zelden delende cellen, zoals B1-NSCs zijn grotendeels rustig en hun naburige ependymale cellen nooit verdelen onder fysiologische omstandigheden 3 vast te stellen. Hier laten we zien dat lentivirale vectoren (LVs) zorgen voor high-efficiency-gen marking en langdurige wijziging van volwassen NSCs en niet-delende ependymale cellen, vanwege meest redelijk om hun vermogen te transduceren en te integreren in het genoom van de doelcellen in een celcyclus-onafhankelijke manier. Verder tonen we aan hoe de route van aflevering en virale titer hulp specifiek transduceren ependymale cellen, maar niet B1 cellen waardoor de analyse van niche-afhankelijke, ependymale effecten op NSC toestaan.

Relatief rustige somatische stamcellen ondersteunen levenslange celvernieuwing in de meeste volwassen weefsels. Neurale stamcellen in de hersenen volwassen zoogdieren zijn beperkt tot twee specifieke neurogene niches: de subgranulaire zone van de dentate gyrus in de hippocampus en de ventriculaire-subventriculaire zone (V-SVZ, ook wel subependymal zone of SEZ) in de wanden van de laterale ventrikels. De ontwikkeling van in vivo genoverdracht-strategieën voor volwassen stamcellen populaties (dwz die van de hersenen van zoogdieren) resulterend in lange termijn expressie van transgenen in de gewenste stamcellen en hun nakomelingen afkomstig is een cruciale rol spelen bij huidig ​​biomedisch en biotechnologisch onderzoek. Hier wordt een directe in vivo methode gepresenteerd voor de stabiele genetische modificatie van volwassen muizen V-SVZ cellen die gebruik maakt van de celcyclus-onafhankelijke infectie door LVs en de zeer gespecialiseerde cytoarchitecture van de V-SVZ niche. LV-gemedieerde genafgifte kan worden gebruikt voor de lange termijn in vivo transductie van cellen in volwassen muizen V-SVZ, waardoor hun tracking en genetische modificatie tijdens proliferatie, migratie en differentiatie. De infectie en de expressie zijn zeer effectief en geven talrijke cellen die gemakkelijk onderscheiden onder andere niet-geïnfecteerde cellen kunnen worden door de expressie van het opgenomen.
