Epithelialen Ovarialkarzinom (EOC) ist die häufigste Todesursache von gynäkologischen Krebserkrankung, mit einem geschätzten 21.290 Neuerkrankungen in den USA im Jahr 2015 und schätzungsweise 14.180 Todesfälle 1. Die überwiegende Mehrheit (&gt; 75%) der Frauen mit Spätstadium der Krankheit diagnostiziert (Stadium III oder IV) durch diffuse Intraperitonealtest Metastasierung und schlechter Prognose charakterisiert. Wiederauftreten der Krankheit in der Bauchhöhle folgenden First-Line - Chemotherapie ist auch üblich , und stellt eine der Hauptursachen für die Sterblichkeit 2,3. EOC metastasiert durch einen einzigartigen Mechanismus sowohl direkte Verlängerung aus dem Primärtumor zu benachbarten Peritonealdialyse Organe sowie durch die Dissoziation oder vergießen von Zellen aus der primären Tumoroberfläche als einzelne Zellen oder mehrzelligen Aggregate beteiligt sind. Die Zellen werden in die Bauchhöhle Schuppen, wobei sie Ablösung induzierte Apoptose 4 widerstehen. Anhaftungen von Peritonealdialyse Aszites ist weit verbreitet, wie Schuppen Tumorzellen Peritonealdialyse Lymphdrainage und t blockierenumors produzieren Wachstumsfaktoren, die die vaskuläre Permeabilität zu verändern. Als primäreWebsite von Eierstockkrebsinzidenz und Metastasierung ist die Bauchhöhle, orthotopen Modelle von EOC Metastasierung die Analyse und Charakterisierung von intraperitoneale Krankheit betreffen. Obwohl es in der Fähigkeit, Bildtumorzellen jüngsten Verbesserungen gewesen, auch auf Einzelzellebene, gibt es immer noch erhebliche Schwierigkeiten in der metastasierten Tumorbelastung von EOC zu quantifizieren. Wir beschreiben hier eine optische Bildgebungsverfahren für die quantitative Analyse von Metastasen eine syngene orthotoper Xenograftmodell von rot fluoreszierendes Protein (RFP) umfasste mit -markierte murine ID8 Ovarkrebszellen 13 und immunkompetenten C57 / BL6 - Mäusen. Wir zeigen , ein neues Verfahren der relativen Quantifizierung der Tumorlast in vivo Vereinigen und ex vivo - Bildgebung mit Entfernung von Gewebe Autofluoreszenz. quantitation of intra-peritoneal ovarian cancer metastasis

Epithelial ovarian cancer (EOC) is the leading cause of death from gynecologic malignancy in the United States. Mortality is due to diagnosis of 75% of women with late stage disease, when metastasis is already present. EOC is characterized by diffuse and widely disseminated intra-peritoneal metastasis. Cells shed from the primary tumor anchor in the mesothelium that lines the peritoneal cavity as well as in the omentum, resulting in multi-focal metastasis, often in the presence of peritoneal ascites. Efforts in our laboratory are directed at a more detailed understanding of factors that regulate EOC metastatic success. However, quantifying metastatic tumor burden represents a significant technical challenge due to the large number, small size and broad distribution of lesions throughout the peritoneum. Herein we describe a method for analysis of EOC metastasis using cells labeled with red fluorescent protein (RFP) coupled with in vivo multispectral imaging. Following intra-peritoneal injection of RFP-labelled tumor cells, mice are imaged weekly until time of sacrifice. At this time, the peritoneal cavity is surgically exposed and organs are imaged in situ. Dissected organs are then placed on a labeled transparent template and imaged ex vivo. Removal of tissue auto-fluorescence during image processing using multispectral unmixing enables accurate quantitation of relative tumor burden. This method has utility in a variety of applications including therapeutic studies to evaluate compounds that may inhibit metastasis and thereby improve overall survival.
