A entrada do vírus influenza A (IAV) é um processo multi-etapas que começa com a ligação do vírus a receptores na membrana plasmática das células alvo 1. O receptor para IAV é ácido siálico que está presente em uma grande variedade de glicoproteínas e glicolípidos. A hemaglutinina (HA) de IAV proteína que está presente no envelope viral se liga ao ácido siálico e assim medeia a ligação das partículas virais na membrana plasmática das células alvo 2. O vírus entra nas células através de endocitose mediada por clatrina, mas também as vias de entrada alternativos, tais como macropinocitose, foram descritos 3-6. A interacção entre o HA e o ácido siálico parece ser suficiente para mediar os dois, a ligação e internalização de disparo de partículas virais 7. No entanto, os receptores de entrada alternativos foram propostos e seus papéis na entrada IAV estão sendo investigados 1,8,9. Medindo diferentes fases de entrada é um desafio IAV experimentalmente como geralmente grandes quantidades de vírus são necessárias para detectar o vírus de entrada. Além disso, a gama de detecção de mudanças na entrada do vírus é única linear devido à falta de replicação viral. Isso ressalta a necessidade de ensaios com alta sensibilidade. O ensaio apresentamos aqui permite a detecção de vírus ligados à superfície da célula, bem como a detecção de vírus internalizado em relação à quantidade total de vírus associados a células. A utilização de vírus de tipo selvagem biotinilado permite a medição conveniente através de coloração com a STV-Cy3 e leitura por citometria de fluxo. Vírus Biotinilado é frio-limite para celulars para permitir a ligação, mas impedir a internalização de partículas virais. As células podem ser fixadas, permeabilizadas e coradas com STV-Cy3 para medir vírus anexado. O sinal de viriões extracelulares, podem ser ligados revogada se um passo de bloqueamento é aplicada antes da fixação, em que as células são incubadas com estreptavidina não marcado (STV). Attachment to target cells followed by internalization are the very first steps of the life cycle of influenza A virus (IAV). We provide here a detailed protocol for measuring relative changes in the amount of viral particles that attach to A549 cells, a human lung epithelial cell line, as well as in the amount of particles that are internalized into the cell. We use biotinylated virus which can be easily detected following staining with Cy3-labeled streptavidin (STV-Cy3). We describe the growth, purification and biotinylation of A/WSN/33, a widely used IAV laboratory strain. Cold-bound biotinylated IAV particles on A549 cells are stained with STV-Cy3 and measured using flow cytometry. To investigate uptake of viral particles, cold-bound virus is allowed to internalize at 37 &#176;C. In order to differentiate between external and internalized viral particles, a blocking step is applied: Free binding spots on the biotin of attached virus on the cell surface are bound by unlabeled streptavidin (STV). Subsequent cell permeabilization and staining with STV-Cy3 then enables detection of internalized viral particles. We present a calculation to determine the relative amount of internalized virus. This assay is suitable to measure effects of drug-treatments or other manipulations on attachment or internalization of IAV.
