high-resolution quantification of odor-guided behavior in drosophila melanogaster using the flywalk paradigm

In hun natuurlijke omgeving, insecten zoals de azijn Drosophila melanogaster worden gebombardeerd met een enorme hoeveelheid chemisch verschillende geurstoffen. Om de zaak nog verder te compliceren, de gedetecteerd door het insect zenuwstelsel geuren zijn meestal niet enkel verbindingen, maar mengsels waarvan de samenstelling en de concentratie ratio&#39;s variëren. Dit leidt tot een bijna oneindig aantal verschillende olfactorische stimuli die moeten worden geëvalueerd door het zenuwstelsel. Om te begrijpen welke aspecten van een geur stimulus zijn bepalend voor de evaluatie door de vlieg, is het daarom wenselijk om efficiënt te onderzoeken geur geleid gedrag ten opzichte van vele geurstoffen en geur mengsels. Om direct correleren gedrag neuronale activiteit, moet het gedrag worden gekwantificeerd in een vergelijkbare tijd en onder dezelfde voorwaarden als stimulans in neurofysiologische experimenten. Echter, veel momenteel gebruikt olfactorische bioassays in Drosophila neuroethology nogal speciaseerd, hetzij in de richting van de efficiëntie of naar resolutie. Flywalk, een geautomatiseerde geur levering en tracking systeem, overbrugt de kloof tussen efficiëntie en resolutie. Zij maakt de bepaling van precies wanneer een geur packet stimuleerde vrijlopende vlieg en de animal&#39;s bepalen dynamische gedragsreactie.

Omdat vliegen zich vrij binnen de glazen buizen verdelen geur pulsen en de geur puls loopt door de glazen buizen met een constante snelheid vliegt ondervindt de geur op verschillende tijdstippen, afhankelijk van hun x-positie ten tijde van de stimulatie. Hierdoor worden de aanzetten van de wind trajecten opgeroepen door een 500 msec puls van een aantrekkelijke 10 -3 verdunning ethylacetaat vertraging van ongeveer 1 sec voor vliegen aan het einde van de wind glasbuizen vergeleken met die van vliegen zitten dic...
