computed tomography and optical imaging of osteogenesis-angiogenesis coupling to assess integration of cranial bone autografts and allografts

En stor parameter bestämma framgången för en ben ympning förfarande är vaskularisering av området kring implantatet. Vi antog att implantation av ett ben autograft skulle få större benåterbyggnad av yppig blodkärlsbildning. För att undersöka effekten av transplantatet på neovaskularisation på skadestället, har vi utvecklat en mikro datortomografi (μCT) tillvägagångssätt för att karakterisera nybildade blodkärl, vilket innebär systemisk perfusion av djuret med en polymeriserande kontrastmedel. Denna metod möjliggör detaljerad vaskulär analys av ett organ i sin helhet. Dessutom var blodperfusion bedömdes med användning fluorescensavbildning (FLI) av en blodburen fluorescerande medlet. Benbildning kvantifierades genom FLI användning av en hydroxiapatit-riktad sond och μCT analys. Stamcell rekrytering övervakades genom bioluminescens avbildning (BLI) av transgena möss som uttrycker luciferas under kontroll av den osteokalcin promotorn.Här beskriver vi och visar framställningen av allograft, calvarial defekt kirurgi, μCT skanningsprotokoll för kärlnybildning studien och benbildning analys (inklusive perfusion av kontrastmedlet in vivo), och protokollet för dataanalys. Den högupplösta analys av kärl 3D uppvisade signifikant större angiogenes hos djur med implanterade auto, särskilt när det gäller arteriole bildning. Följaktligen blodperfusion var signifikant högre i gruppen med autologt bentransplantat från 7: e dagen efter operationen. Vi observerade överlägsen benmineralisering och mäts större benbildning hos djur som fick auto. Autograft implantering inducerad bosatta stamcells rekrytering till transplantat-värdbenet sutur, där cellerna differentieras till benbildande celler mellan den 7: e och 10: e postoperativa dagen. Detta konstaterande innebär att förbättrad formation ben kan tillskrivasden utökade kärl utfodring som präglar autograft implantation. De metoder som avbildas kan tjäna som ett optimalt verktyg för att studera benåterbyggnad i form av tätt avgränsas benbildning och kärlnybildning.

Neovaskularisation bedömdes av μCT volymetriska analyser och FLI hjälp av en fluorescerande blodburen agent för att kvantifiera blodperfusionen. Sju dagar efter operationen, μCT scanning visade en betydligt högre volym av små och medel diameter blodkärl hos möss som hade fått auto än hos möss som hade fått allografter skördas från C57BL / 6 (figur 3A).
