Les hydrogels sont particulièrement prometteurs biomatériaux, et devraient être importantes en biologie fondamentale, essais pharmacologiques et de la médecine. 1 Biofabrication de constructions cellulaires à base hydrogel a été suggéré de réduire l&#39;utilisation de l&#39;expérimentation animale, 2,3 remplacer des tissus transplantables, 4 et améliorer tests cellulaires. 5,6 contenant de l&#39;eau-hydrocarbures) (matériaux viscoélastiques (gels) permettent à un grand nombre de cellules à encapsuler et maintenu dans une structure d&#39;échafaudage pour contrôler le microenvironnement cellulaire 3D. En combinaison avec la direction des technologies microfluidiques ou micromodelage, la géométrie des constructions d&#39;hydrogel peut être contrôlée avec précision à l&#39;échelle cellulaire. Les deux particules et fibres à base hydrogel ont été facilement et rapidement fabriqués pour des applications comme les environnements cellulaires micro-échelle, avec des commandes fluidiques utilisant des dispositifs microfluidiques. La technique permet micromodelage complexe et de la conception modulaire des constructions cellulaires par l&#39;intermédiaire d&#39;un processus d&#39;empilement de structures multicouches. Grâce à la fabrication de feuilles empilées d&#39;hydrogel modulaires, qui sont microélectrodes, un système de culture cellulaire 3D avec un microenvironnement cellulaire macro-échelle contrôlée peut être réalisée. Ce protocole de vidéo décrit un procédé simple mais puissant de fabrication qui peut être utilisé pour construire des feuilles d&#39;hydrogel modulaires, basé sur la lignée cellulaire de carcinome de foie humain (HepG2). Hydrogels peuvent être modelés à la micro-échelle en utilisant des technologies microfluidiques ou micromodelage de fournir une vivo -comme géométrie en trois dimensions (3D) des tissus. Les constructions 3D résultant base d&#39;hydrogel-cellulaires ont été introduits comme une alternative à l&#39;expérimentation animale pour les études biologiques de pointe, essais pharmacologiques et les applications de transplantation d&#39;organes. Bien que les particules et fibres à base hydrogel peuvent être facilement fabriqués, il est difficile de les manipuler pour la reconstruction tissulaire. Dans cette vidéo, nous décrivons un procédé de fabrication de feuilles d&#39;hydrogel d&#39;alginate microélectrodes, ainsi que leur assemblage pour former un système de culture cellulaire 3D macro-échelle avec un microenvironnement cellulaire contrôlée. Utilisation d&#39;une forme de brouillard de l&#39;agent gélifiant de calcium, de minces feuilles d&#39;hydrogel peuvent être facilement générées avec une épaisseur dans la plage de 100 - 200 pm, et avec micropatterns précises. Les cellules peuvent ensuite être cultivées à l&#39;orientation géométrique des feuilles d&#39;hydrogel dansconditions autonome. En outre, les feuilles d&#39;hydrogel peuvent être facilement manipulées en utilisant une micropipette avec une extrémité de bout en coupe, et peuvent être assemblés en structures multicouches en les empilant l&#39;aide d&#39;un polydiméthylsiloxane à motifs (PDMS) de trame.
