La pérdida completa de los cilios en los resultados en desarrollo cóclea en conductos cocleares acortar gravemente, mal formado y mal orientada haces estereociliar, así como los cuerpos basales mis-posicionado 8,9. Un cilio funcional no es sólo formaba parte del axonema ciliar. Muchas proteínas asociadas con los ciliosse producen en función de los complejos localizados en subdominios cilios afines, como la basal del cuerpo, zona de transición, o axonema ciliar 15. El cuerpo básica, derivada del centríolo madre del centrosoma, es también un centro organizador de microtúbulos por los microtúbulos se extienden lejos del cilio en el cuerpo celular y puede regular el tráfico intracelular, así como el tráfico ciliar. La zona de transición ciliar es otra región donde la función ciliar se regula en función de la organización de importación y exportación de compuestos ciliares 16. Múltiples estudios han identificado una conexión entre los cilios y Wnt no canónica (de señalización PCP), aunque el mecanismo exacto no está claro 17. La redundancia de ciliares y PCP genes y la sensibilidad de la polaridad celular a anormalidades celulares generalizadas, hacen que sea difícil para enlazar directamente a una mutación específica déficit de PCF. Una de las salidas de lectura de la señalización de PCP es el posicionamiento del cuerpo basal y primarcilio y, por lo tanto, la segregación de la primaria de los defectos secundarios es un reto. Algunos estudios en el pez cebra y el ratón mutantes han sugerido ninguna conexión entre cilios y la señalización Wnt 18-20. Las discrepancias en los datos siempre son representativas de las especies, tejidos, o las diferencias temporales que dependen de las contribuciones ciliares hacia la señalización de Wnt. Además, la capacidad de respuesta normal de Wnt podría ser retenida si cuerpos basales siguen siendo funcionales. Una comprensión más profunda de la función de las proteínas ciliares en vías de señalización celular y otros fenómenos biológicos es crucial para nuestra comprensión de la biología celular y del desarrollo, así como para el desarrollo de estrategias de tratamiento dirigidas.

evaluation of planar-cell-polarity phenotypes in ciliopathy mouse mutant cochlea

In recent years, primary cilia have emerged as key regulators in development and disease by influencing numerous signaling pathways. One of the earliest signaling pathways shown to be associated with ciliary function was the non-canonical Wnt signaling pathway, also referred to as planar cell polarity (PCP) signaling. One of the best places in which to study the effects of planar cell polarity (PCP) signaling during vertebrate development is the mammalian cochlea. PCP signaling disruption in the mouse cochlea disrupts cochlear outgrowth, cellular patterning and hair cell orientation, all of which are affected by cilia dysfunction. The goal of this protocol is to describe the analysis of PCP signaling in the developing mammalian cochlea via phenotypic analysis, immunohistochemistry and scanning electron microscopy. Defects in convergence and extension are manifested as a shortening of the cochlear duct and/or changes in cellular patterning, which can be quantified following dissection from developing mouse mutants. Changes in stereociliary bundle orientation and kinocilia length or positioning can be observed and quantitated using either immunofluorescence or scanning electron microscopy (SEM). A deeper insight into the role of ciliary proteins in cellular signaling pathways and other biological phenomena is crucial for our understanding of cellular and developmental biology, as well as for the development of targeted treatment strategies.
