Die überwiegende Mehrheit der Massenspektrometrie (MS) -Basis Proteinanalyseverfahren beinhalten ein enzymatisches Verdauungsschritt der Detektion vor, typischerweise mit Trypsin. Dieser Schritt ist für die Erzeugung von niedermolekularen Peptide erforderlich, in der Regel mit MW &lt;3000-4000 Da, die innerhalb des effektiven Scan-Bereich der Massenspektrometrie Instrumentierung fallen. Herkömmliche Protokolle beinhalten O / N enzymatische Verdauung bei 37 ºC. Jüngste Fortschritte haben zu der Entwicklung einer Vielzahl von Strategien geführt, in der Regel die Verwendung eines Mikroreaktor mit immobilisierten Enzymen oder aus einer Reihe von komplementären physikalischen Prozessen beteiligt, die die Zeit , die für proteolytischen Verdau zu einigen Minuten (beispielsweise Mikrowellen- oder Hoch reduzieren Druck). In dieser Arbeit beschreiben wir eine einfache und kostengünstige Lösung, die in einem Labor für die Erreichung schnelle enzymatische Verdauung eines Proteins umgesetzt werden können. Das Protein (oder Proteingemisch) adsorbiert an C18-gebundene Umkehrphasen-Hochleistungormance Flüssigchromatographie (HPLC) Siliciumdioxidteilchen in einer Kapillarsäule vorgespannt, und Trypsin in wässrigem Puffer wird für eine kurze Zeit über die Teilchen infundiert. Zu ermöglichen Online-MS-Detektion, die tryptischen Peptide werden mit einem Lösungsmittelsystem mit einem erhöhten Gehalt an organischen Stoffen direkt in der MS-Ionenquelle eluiert. Dieser Ansatz vermeidet die Verwendung von hochpreisigen immobilisiertem Enzympartikel und erforderlich macht keine Hilfe für den Prozess abzuschließen. Protein Verdauung und vollständige Probenanalyse kann in weniger als ~ 3 min und ca. 30 min bzw. erreicht werden.
