Ce protocole décrit la méthode utilisée pour isoler et classer l&#39;antre de la souris ovocytes GV. Il n&#39;y a pas des étapes cruciales notamment pour souligner, à quelques exceptions près. Premièrement: cette procédure doit être effectuée aussi rapidement que possible à: a) maintenir la vitalité des ovocytes et b) d&#39;éviter les changements de pH dans le moyen utilisé. Antraux ovocytes entièrement développés (GV, vésicule germinale) isolés du compartiment de l&#39;antre de l&#39;ovaire sont couramment utilisés à des fins différentes, tels que, par exemple, l&#39;étude des mécanismes moléculaires et cellulaires derrière la maturation in vitro jusqu&#39;au stade métaphase II. Malgré leurs belles apparence ovocytes plus antre, mais pas tous, sont en mesure de compléter la méiose et atteindre le stade de blastocyste; En fait, dans de nombreuses espèces de mammifères, dont les humains 1,2, environ 30% d&#39;entre eux arrêter leur développement au stade 2 cellules, si la fécondation se produit 3-5. À ce jour, peu de paramètres sont utilisés pour distinguer entre les ovocytes capables de soutenir le développement embryonnaire de ceux qui sont incapables de le faire. Une autre méthode bien établie réside dans leur organisation de la chromatine: si colorées avec des colorants intercalants de l&#39;ADN (comme Hoechst33342), 70% des ovocytes antraux montre un anneau de colorant positive autour du nucléole et sont appelés Entouré Nucléole (SN), tandis que les 30% restants montrer un des signaux positifs plus tachetées et sont appelés pas entouré Nucléole (NSN) 3-5. Récemment, nous avons montré que l&#39;absence de réseaux cytoplasmiques 9 (de CPL, sites de stockage importants pour l&#39;ARNm et des ribosomes d&#39;origine maternelle dans les deux ovocytes et des embryons de mammifères) 10 et la régulation à la baisse de MATER (essentiel pour la formation CPL 11), avec la présence de gouttelettes de lipides dans leur cytoplasme 9,12, peuvent être d&#39;autres causes liées à l&#39;incapacité des ovocytes NSN à aller au-delà du stade 2 cellules. Malheureusement, quelques techniques peuvent être appliquées pour trier SN antre et ovocytes RSN et,pour cette raison, le développement d&#39;une méthode moins invasive (sans coloration nucléaire, les procédures de fixation ou de manipulation aux pipettes bouche) pourrait devenir très important d&#39;augmenter les taux de développement de l&#39;embryon à la fois humaine et animale. Dans cet article, nous détaillons le protocole simple et rapide utilisé pour isoler annonce SN de tri et RSN ovocytes antre de souris femelles et elle est adressée à tous les scientifiques intéressés par l&#39;étude de la gamétogenèse femelle, la maturation des ovocytes et le développement de l&#39;embryon.

isolation and characterization of mouse antral oocytes based on nucleolar chromatin organization

This protocol describes a simple and quick method to isolate and characterize mouse antral GV (Germinal Vesicle) oocytes as able (SN, Surrounded Nucleolus) or unable (NSN, Not Surrounded Nucleolus) to develop to the blastocyst stage after in vitro maturation (IVM) and in vitro fertilization (IVF). It makes use of Hoeschst33342 (or any other DNA intercalating dye) able to bind to the heterochromatin of the nucleolus showing a ring in the SN oocytes or not, like in the NSN oocytes. This represents the easiest and quickest way to sort both antral oocytes that can be eventually used for IVM or IVF procedures. 
Briefly, the protocol consists of the following steps: hormone injection to stimulate follicular growth; isolation of the oocytes at the GV stage from the antral compartment by puncturing the ovary with a sterile needle; preparation of thin glass pipettes for mouth pipetting of the oocytes; sorting of the oocytes with Hoechst33342 prepared at a supravital concentration; IVM, IVF or any other molecular/cellular analysis.
Unfortunately there are still few evidences to sort SN and NSN oocytes using less invasive techniques. If and once they will be identified, they could be potentially applied to human assisted reproductive technologies, although with several aspects that should be modified. To date, this technique has potential implications to dramatically increase IVM and IVF successful procedures in both endangered and species with economic interest.
