Characterizing förändringar i huden hemmahörande leukocyter i gnagarmodeller av hudsjukdomar som atopisk dermatit eller psoriasis är viktigt för att förstå mekanistiska kopplingar mellan inflammatoriska celler tillströmning och sjukdomspatologin. Hudåkommor som sträcker sig från kontakteksem, eksem, psoriasis, cellulit, svampinfektioner och bölder till icke-melanom hudcancer befanns vara bland de 50 vanligaste sjukdomarna i världen, och den fjärde ledande orsaken till icke-dödliga sjukdomar under 2010 1. Följaktligen är utredningen av molekylära och cellulära mekanismer som ligger bakom olika hud sjukdomar en nödvändig och aktivt forskningsområde. Gnagarmodeller har varit anmärkningsvärt användbar vid förståelsen av inflammatoriska hudtillstånd såsom atopisk dermatit 2, psoriasis 3, eller Staphylococcus aureus-infektion 4. Inexpensive, effektiva och enkla protokoll för enzymatisk digerering av mus hudvävnad kan tillhandahålla beredningar av celler som kan användas för en mängd olika tillämpningar nedströms för att bättre förstå patofysiologin av hudsjukdomar. Här används en enkel och ekonomisk metod som beskrivits för enzymatisk uppslutning av mushudvävnad och isolering av huden infiltrerande leukocyter som kan användas för cellodling in vivo adoptiv överföring, flödescytometrisk analys och sortering eller gen expressionsstudier. Det övergripande målet med detta förfarande är att förbereda en enda cell suspension av hud-infiltrerande leukocyter med hög cellviabiliteten samtidigt minimera kostnaderna vanligtvis förknippas med anpassade reagenskit och mekaniska dissociators. Befintliga hud vävnadsdissociering metoder 5-7 kan leda till låg cellviabilitet och ytmarkör integritet, eller kräva anpassade enzymsatser och dyra vävnadsdissociering maskiner 8-11. Även rötning av musöra hudvävnad är tämligen utbredd 12-13, smälta mycket keratiniserad hudvävnad (t.ex. från flanken) kan resultera i cellpreparat kontaminerade med stora mängder av icke-cellulärt skräp. Andra protokoll använder RPMI 1640 i matsmältningen mediet 6. Emellertid kan närvaron av Mg2 + och Ca 2 + i RPMI orsaka omfattande cellaggregation 17. En idealisk protokoll för vävnadsdissociering bör sträva efter hög cellviabilitet, lågnivåer av cellaggregering och minimal skada på cellyteproteiner. Hög kvalitet preparat lymfkörtel stromacellinjer har åstadkommits med protokoll som använder kortare enzym inkubationer, Ca2 + och Mg 2 + fria medier, och undvika trypsin och dispas 18. Emellertid har protokoll av detta slag inte fastställts för dissociation av hela mushud. isolation of infiltrating leukocytes from mouse skin using enzymatic digest and gradient separation

Dissociating murine skin into a single cell suspension is essential for downstream cellular analysis such as the characterization of infiltrating immune cells in rodent models of skin inflammation. Here, we describe a protocol for the digestion of mouse skin in a nutrient-rich solution with collagenase D, followed by separation of hematopoietic cells using a discontinuous density gradient. Cells thus obtained can be used for in vitro studies, in vivo transfer, and other downstream cellular and molecular analyses including flow cytometry. This protocol is an effective and economical alternative to expensive mechanical dissociators, specialized separation columns, and harsher trypsin- and dispase-based digestion methods, which may compromise cellular viability or density of surface proteins relevant for phenotypic characterization or cellular function. As shown here in our representative data, this protocol produced highly viable cells, contained specific immune cell subsets, and had no effect on integrity of common surface marker proteins used in flow cytometric analysis.
