Stromingspatroon ontwerp is de eerste cruciale stap in het vervaardigen van de 2-D microarray. Antilichaam microarrays zijn op grote schaal gebruikt in proteomics studies decennia de aanwezigheid van gerichte eiwitten, waaronder eiwitten biomarkers 1-3 onderzocht. Hoewel dit gebied die momenteel wordt geconfronteerd met grote uitdagingen van andere high-throughput technologieën zoals massaspectrometrie (MS), is er nog voldoende ruimte voor het nut van antilichaam microarrays, vooral omdat deze apparaten veroorloven eenvoudige interpretatie van de gegevens en gemakkelijke interface met andere testen. De afgelopen jaren is de integratie van microarrays in microchip scaffolds mits het antilichaam microarray een nieuwe kans te gedijen 4-7. Zo heeft de barcode microarray geïntegreerd in een single-cell chip is gebruikt in mobiele communicatie studies 8,9. Deze technologie heeft onderscheidende voordelen boven andere beschikbare microarray technologie. Het beschikt arrayelementen op 10-100 um, veel kleiner dan de typische 150 urn size in gebruikelijke microarray elements. De constructie van kleinere arrayelementen wordt bereikt door systematisch flow patronen benaderingen, en dit geeft aanleiding tot compact microarrays die eencellige uitgescheiden eiwitten en intracellulaire eiwitten kan detecteren. Een ander voordeel is het gebruik van een eenvoudige, instrument set-up. Dit is vooral belangrijk, omdat de meeste laboratoria en kleine bedrijven kunnen niet in staat zijn om toegang te krijgen tot microarray kernfaciliteiten. Dergelijke barcode antilichaam microarrays uitgevoerd met verbeterde doorvoer assay en kunnen worden gebruikt om zeer multiplex assays op enkele cellen terwijl het bereiken van een hoge gevoeligheid en specificiteit vergelijkbaar met die van conventionele sandwich enzyme-linked immunosorbent assay (ELISA 8). Deze technologie heeft talrijke toepassingen gevonden in het opsporen van eiwitten uit glioblastoma 9-11, T-cellen 12 en circulerende tumorcellen 13. Als alternatief, barcode DNA microarrays alleen zijn gebruikt in de nauwkeurige positionering van neuronen en astrocyten voor nabootsening de in vivo assemblage van hersenweefsel 14. Dit protocol alleen gericht op de experimentele stappen en opbouw blokken van de tweedimensionale (2D) streepjescode antilichaam microarray die potentiële toepassingen bij de detectie van biomarkers in vloeibare monsters heeft en in enkele cellen. De technologie is gebaseerd op adresseerbare enkelstrengig, eendimensionale (1-D) DNA microarray geconstrueerd met oligonucleotiden die orthogonaal ruimtelijk patroon op glassubstraten. De 1-D wordt gevormd als evenwijdige stromingskanalen worden toegepast in de stroom-patroonvormingsstap en dergelijk patroon weergegeven als discrete banden visueel vergelijkbaar 1-D Universal Product Code (UPC) barcodes. De constructie van een 2-D (n x m) antilichaam matrix - doet denken aan een 2-D Quick Response (QR) matrixcode - dient complexer patroon strategieën, maar maakt de immobilisatie van antilichamen met een hogere dichtheid 8,15. De fabricagevereist twee DNA patroonvormende stappen, de eerste patroon loodrecht op de tweede. De snijpunten van deze twee patronen vormen de n x m elementen van de array. Door strategische keuze van de reeksen van enkelstrengs DNA (ssDNA) gebruikt in flow-patroonvorming, is elk element in een bepaalde rij een specifiek adres toegewezen. Deze ruimtelijke referentie is noodzakelijk onderscheid tussen fluorescentiesignalen op de microarray dia. De ssDNA matrix wordt omgezet in een antilichaam matrix door de integratie van complementaire DNA-antilichaam conjugaten, waarbij een platform genaamd DNA gecodeerde bibliotheek antilichaam (DEAL 16). Deze video protocol beschrijft de belangrijkste stappen in het creëren van nxm antilichaam-arrays, die onder meer het opstellen van polydimethylsiloxaan (PDMS) barcode mallen, stroom-patroonvorming ssDNA in twee oriëntaties, het voorbereiden van antilichaam-oligonucleotide DEAL conjugaten, en het omzetten van de 3 x 3 DNA-array in een 3x 3 antilichaam array.

flow-pattern guided fabrication of high-density barcode antibody microarray

Antibody microarray as a well-developed technology is currently challenged by a few other established or emerging high-throughput technologies. In this report, we renovate the antibody microarray technology by using a novel approach for manufacturing and by introducing new features. The fabrication of our high-density antibody microarray is accomplished through perpendicularly oriented flow-patterning of single stranded DNAs and subsequent conversion mediated by DNA-antibody conjugates. This protocol outlines the critical steps in flow-patterning DNA, producing and purifying DNA-antibody conjugates, and assessing the quality of the fabricated microarray. The uniformity and sensitivity are comparable with conventional microarrays, while our microarray fabrication does not require the assistance of an array printer and can be performed in most research laboratories. The other major advantage is that the size of our microarray units is 10 times smaller than that of printed arrays, offering the unique capability of analyzing functional proteins from single cells when interfacing with generic microchip designs. This barcode technology can be widely employed in biomarker detection, cell signaling studies, tissue engineering, and a variety of clinical applications.
