in situ characterization of hydrated proteins in water by salvi and tof-sims

Dit werk toont in situ karakterisering van eiwit biomoleculen in de waterige oplossing met behulp van het systeem voor de analyse van de Liquid Vacuum Interface (SALVI) en time-of-flight secundaire ionen massaspectrometrie (ToF-SIMS). De fibronectine proteïnefilm werd geïmmobiliseerd op het siliciumnitride (SiN) membraan dat de SALVI detectiegebied vormt. Tijdens ToF-SIMS werden drie wijzen van analyse zoals hoge ruimtelijke resolutie massaspectrometrie, tweedimensionale (2D) beeldvorming en diepteprofilering. Massaspectra werden zowel positieve als negatieve modes verworven. Gedeïoniseerd water werd ook geanalyseerd als referentiemonster. Onze resultaten tonen aan dat de film fibronectine in water is duidelijker en sterker watercluster pieken opzichte water alleen. Karakteristieke pieken aminozuur fragmenten zijn ook waargenomen in het gehydrateerde eiwit ToF-SIMS spectra. Deze resultaten illustreren dat eiwitmolecuul adsorptie aan een oppervlak kan worden bestudeerd dynamically gebruik SALVI en ToF-SIMS in de vloeibare omgeving voor de eerste keer.

Enkele representatieve resultaten worden de voordelen van de voorgestelde protocol aangetoond. Door het gebruik van de SALVI microfluïdische interface kan de primaire ionenbundel (Bi 3 +) direct bombarderen op de gehydrateerde fibronectine film in DI water.
