in situ characterization of hydrated proteins in water by salvi and tof-sims

Diese Arbeit zeigt, in-situ-Charakterisierung von Protein Biomolekülen in der wässrigen Lösung in dem Liquid Vacuum-Schnittstelle des Systems für die Analyse unter Verwendung von (SALVI) und Time-of-Flight-Sekundärionenmassenspektrometrie (TOF-SIMS). Die Fibronektin Proteinfilm wurde auf dem Siliziumnitrid (SiN), die die Membran immobilisiert SALVI Erfassungsbereich bildet. Während ToF-SIMS-Analyse wurden drei Arten der Analyse durchgeführt, einschließlich hoher räumlicher Auflösung Massenspektrometrie, zweidimensionale (2D) Bildgebung und Tiefenprofilierung. Massenspektren wurden im positiven und negativen Modus erworben. Entionisiertes Wasser wurde als Referenzprobe analysiert. Unsere Ergebnisse zeigen, dass die Fibronektin Film in Wasser hat deutlicher und stärker Spitzen Wassercluster zu Wasser allein verglichen. Charakteristische Peaks von Aminosäure-Fragmente sind ebenfalls zu beobachten in der hydratisierten Protein TOF-SIMS-Spektren. Diese Ergebnisse zeigen, dass Proteinmolekül Adsorption an einer Oberfläche dynamica sucht werden könnenlly Verwendung SALVI und ToF-SIMS in der flüssigen Umgebung zum ersten Mal.

Einige repräsentative Ergebnisse sind die Vorteile des vorgeschlagenen Protokolls zu demonstrieren. Durch die Verwendung der Mikrofluid-SALVI Schnittstelle der Primärionenstrahl (Bi 3 +) auf dem hydratisierten Film Fibronektin in DI Wasser direkt bombardieren.
