in situ characterization of hydrated proteins in water by salvi and tof-sims

עבודה זו ממחישה באפיון באתרו של ביומולקולות חלבון בתמיסה המימית באמצעות מערכת לניתוח בבית ממשק האבק הנוזלי (Salvi) וזמן של הטיסה ספקטרומטריית משני יון המוני (TOF-SIMS). סרט חלבון פיברונקטין מאובן על סיליקון ניטריד (חטא) קרום הנוצר באזור זיהוי Salvi. במהלך ניתוח TOF-SIMS, שלושה מצבים של ניתוח נערכו כולל ספקטרומטריית מסה ברזולוציה מרחבית גבוהה, דו מימדי (2D) הדמיה, ו פרופיל עומק. ספקטרה המונית נרכשה בשני מצבים חיוביים ושליליים. מים ללא יונים נותחו גם כמדגם התייחסות. התוצאות שלנו מראה כי סרט פיברונקטין במים יש יותר פסגות אשכול מים ברורות וחזקות נמשלו למים לבד. פסגות מאפיין של שברי חומצת אמינו ניתנים לצפייה גם בחלבון hydrated ספקטרה TOF-SIMS. תוצאות אלו ממחישות כי ספיחת מולקולת חלבון על משטח ניתן ללמוד דינמיקהlly באמצעות Salvi ו TOF-SIMS בסביבת הנוזל בפעם הראשונה.
