I dette papir rapport vi om en enkel metode til at sammenligne celle justering adfærd efter hydrogel figur vejledning og trækstyrke strækning. Tjener som en blok materiale, der efterligner en native mikromiljø, kan en hydrogel, som indeholder ekstracellulære matrix (ECM) genetablere opbygge biomimetiske væv stilladser til støtte for cellevækst. Til at besidde funktionerne af en væv, er organiseret cellejustering en væsentlig forudsætning. Forskellige 2D (dvs. celler dyrkes på en overflade) og 3D (dvs. celler indkapslet i en hydrogel) celle alignments har opnået ved dyrkning eller indkapsle celler i eller på fleksible substrater med mikro- eller nano-mønstre1. 3D cellejustering i mikroarkitekturen er mere attraktive, da mikromiljø er tættere på den indfødte væv konstruere2,3,4. En almindelig metode til 3D cellejustering er den geometriske cue hydrogel figur2,3. På grund af den begrænsede plads til celleproliferation i kort-akse retningen, celler har til formål at bringe langs lange-akse retningen i en mikro-mønstrede hydrogel. En anden metode er at anvende trækstyrke strækning til hydrogels at opnå cellejustering parallelt med stretch retning4,5.
Biofysiske stimulation på ECM hydrogels, såsom trykstyrke stamme eller et elektrisk felt, kan regulere cellefunktioner for ordentlig væv integration, spredning og differentiering1,2,3. Megen forskning er blevet gjort for at undersøge cellulære adfærd ved at anvende én stamme tilstand ad gangen ved hjælp af flere mekaniske kontrol enheder4,6,7,8,9. For eksempel, brug af mekaniske trin motors klemt eller strakt på en 3D celle-indkapslede kollagen hydrogel har været en fælles tilgang7,10. Men sådanne kontrollerende udstyr kræver ekstra plads og står over for spørgsmålet om forurening i inkubator7,9,11,12. Derudover kan ikke det store instrument give en præcis kontrolmiljø at give høj reproducerbarhed13.
I betragtning af, at celle-laden hydrogels normalt er beskæftiget på mikro-skalaen for biomedicinske programmer, er det en fordel at kombinere MEMS teknikker for at generere en række stamme/stretch stimulation til samtidig undersøge celle adfærd i 3D biomimetiske konstruktioner in vitro-2,14,15,16,17,18. For eksempel kan bruger gas pres til at deformere PDMS membran i mikrofluid chips give anledning til forskellige stammer, køre Celledifferentiering til forskellige slægter9,16. Men der er mange tekniske udfordringer, såsom komplicerede chip fabrication processer i et rent værelse og software kontrol integration af motorer, pumper, ventiler og komprimerede gasser.
I dette arbejde viser vi en enkel tilgang for at opnå en selvbærende gradient statisk-stamme mikrofluid chip ved at ansætte en koncentrisk cirkulære hydrogel mønster og en fleksibel PDMS membran. I modsætning til de fleste af de eksisterende metoder er vores platform en transportabel og engangs miniature enhed der kan fremstilles uden for en gul stue og der besidder selv generere gradient stammer på koncentriske celle-indkapslede hydrogels, uden ydre mekanisk udstyr under inkubation. 3T3 fibroblast celle adfærd påvirkes af en kombination af hydrogel form og en bred vifte af trækstyrke stretch vejledning stikord blev påvist under observation af cellejustering i 3D ECM-mimetiske miljøer i gradient stamme chip for 3 dage.

Kunstige vejledning for cellulære justering er et varmt emne i feltet af vævsmanipulering. De fleste af de tidligere forskning har undersøgt enkelt stamme-induceret cellulære justering på en celle-belæsset hydrogel ved hjælp af komplekse eksperimentelle processer og masse kontrol systemer, som er normalt forbundet med forurening spørgsmål.
