Her beskriver vi en tilpasningaf fluorescensen farvestof fremgangsmåde til en face samtidig detektion og visualisering af intracellulær NO og O 2 .- hjælp cellen permeable diaminofluorescein-2 diacetat (DAF-2DA) og dihydroethidium (DHE), henholdsvis i intakte aorta hos en fedme musemodel inducerede af højt fedtindhold-kost fodring. Vi kunne påvise formindsket intracellulær NO og forbedret O 2 .- niveauer i frisk isolerede intakte aorta hos fedme mus sammenlignet med kontrollen lean mus. Vi viser, at denne metode er en nem teknik til direkte påvisning og visualisering af NO og O 2 .- i de intakte blodkar og kan i vid udstrækning anvendes til undersøgelse af endotel (dys) funktion under (fysio) patologiske tilstande.

Fedme er en vigtig risikofaktor for iskæmisk hjertesygdom og er forbundet med nedsat endotel NO biotilgængelighed, et kendetegn for aterosklerotisk vaskulær sygdom 21. eNOS-frakobling har vist sig at være en vigtig mekanisme af endothelial dysfunktion under mange fysiologiske og patologiske tilstande, herunder ældning 22, aterosklerose og fedme 14.
