Vanwege de uiterst labiele aard van NO en O 2 .-, is moeilijk te meten NO en O 2 .- direct in een bloedvat. Talrijke werkwijzen zijn ontwikkeld voor het meten en NO O 2 .- productie. Het is echter ook ongevoelig of niet-specifieke of technisch veeleisend en vereist speciale apparatuur. Hier beschrijven we een aanpassingvan de fluorescentie kleurstof werkwijze voor het gezicht en gelijktijdige detectie en visualisatie van intracellulaire NO en O 2 .- via de cel permeabele diaminofluorescein 2-diacetaat (DAF-2DA) en dihydroethidium (DHE), respectievelijk in intacte aorta van een obesitas muismodel opgewekt door hoog-vet dieet geeft. Wij konden aantonen verminderde intracellulaire NO en verbeterde O 2 .- niveaus in de vers geïsoleerde intacte aorta van obesitas muis in vergelijking met de controle lean muis. We tonen aan dat deze werkwijze een gemakkelijke techniek voor directe detectie en visualisatie van NO en O 2 .- in de intacte bloedvaten en kan wijd voor onderzoek endotheel (dys) functie onder (fysio) pathologie.
