Heteroxylane sind eine Familie von Polysacchariden , die die vorherrschenden nicht-cellulosischen Polysacchariden der Primärwänden von Gräsern und den sekundären Wänden aller Angiospermen 3-6 sind. Die Xylan - Backbones unterscheiden sich in ihren Typen und Muster der Substitution mit Glykosyl (Glucuronsäure (GlcA), Arabinose (Araf)) und nicht-Glycosyl (O-acetyl, Ferulasäure) -Reste je nach Gewebetyp, Entwicklungsstadium und Art 7. Wände aus Weizen (Triticum aestivum L.) Endosperm bestehen hauptsächlich aus Arabinoxylane (AXs) (70%) und (1 → 3) (1 → 4) zusammengesetzt -β-D-Glucane (20%) mit geringen Mengen an Cellulose und heteromannans (je 2%) 8. Die Xylan-Rückgrat kann verschieden un-substituiert sein und überwiegend mono-substituierte (hauptsächlich O-2-Position und in einem geringeren Ausmaß O-3 Position) und di-substituierte (O-2 und O-3-Positionen) mit α-L-Ara f Reste 9. Das Reduktions Ende (RE) von heteroXylane aus dikotylen Pflanzen (beispielsweise Arabidopsis thaliana) 10 und Gymnospermen (beispielsweise Fichte (Picea abies)) 11 enthält eine charakteristische Tetrasaccharid Glycosyl - Sequenz; -β-D-Xyl p - (1 → 3) -α-L-Rha p - (1 → 2) -α-D-Gal p A- (1 → 4) -D-Xyl p. Um zu verstehen, heteroxylan Biosynthese und Funktion (biologische und Industrie), ist es wichtig, um vollständig die Xylan-Hauptsequenz die Typen und die Muster der Substitutionen sowie die Sequenz des reduzierenden Ende (RE) zu verstehen. Spezifische Techniken zur strukturellen Charakterisierung von reduzierenden Ende (RE) und inneren Bereich Glycosyl-Sequenz (en) von Heteroxylane sind in diesem Manuskript beschrieben. Die Techniken beruhen auf Fluorophore (mit 2 Aminobenzamid (2AB)) das reduzierende Ende (RE) der heteroxylan Kette vor der enzymatischen (Endoxylanase) Hydrolyse Tagging. Dieser Ansatz, insbesondere für die RE Sequenzierung wurdezuerst vom Labor Yorker berichtet 10,12-13 aber jetzt sind der innere Bereich Sequenzierung erweitert und ist eine Kombination von etablierten Techniken, die für alle Heteroxylane unabhängig von ihrer Quelle der Isolation ebenso anpaßbar ist. Dieser Ansatz kann auch auf andere Klassen von Polysacchariden verwendet (soweit verfügbar) die entsprechenden endo-Hydrolasen angewendet werden. In der vorliegenden Studie de-gestärkte wheat endosperm Zellwände wurden als ein in Alkohol unlösliche Rückstand (AIR) und der Reihe nach mit Wasser (W-sol Fr) und 1 M KOH , enthaltend 1% NaBH 4 (KOH-sol Fr) extrahiert wie beschrieben isoliert in Ratnayake et al. (2014) 2. Die freigesetzten Oligosaccharide von beiden W- und KOH-sol Frs dann methyliert und die detaillierte Strukturanalyse von sowohl den nativen und methylierte Oligosaccharide erfolgt mit einer Kombination von MALDI-TOF-MS, ESI-QTOF-MS-Verbindung mit HPLC mit der Verwendung Online chromatographische Trennung einer RP C-18-Säule unter Verwendung vonund ESI-MS n. Endoxylanase verdaut KOH-sol AXs auch durch magnetische Kernresonanz (NMR) charakterisiert wurde.

Dieses Protokoll beschreibt die spezifischen Techniken zur Charakterisierung verwendet Ende Reduktions (RE) und inneren Bereich Glycosyl-Sequenz (en) von Heteroxylane.
