Mesenchymale stromale celler (MSC) er i økende grad anerkjent for deres terapeutiske potensial i et bredt spekter av sykdommer, inkludert lungesykdommer. I tillegg til bruk av benmarg og navlestreng MSC for eksogene celleterapi, er det også økende interesse for reparasjon og regenerativ potensialet bosatt vev MSC. Under utviklingen, blant annet i lungene, er det mesenchyme en viktig kilde til utviklings signaler, og bosatt MSC er en sannsynlig kandidat til å være i sentrum for dette. Disse varierer fra fluorescens-aktivert cellesortering (FACS) baserte metoder som selekterer for CD31 - / CD45 - / CD90 + celler 7, CD31 - / CD45 - / epithelial cell adhesion molecule (EpCAM) - / Sca-1 + -celler til 8, multiresistens transporter ATP bindende kassett G (ABCG2) positive celler 9 eller Hoechst 33342 fargestoff utstrømming 10, til plast etterlevelse 11,12 og migrasjon ut av hakket vev 13. Fordelene med den her presenterte fremgangsmåten er flere ganger. Dette er en fordel i forhold til å bruke CD90 som en seleksjonsmarkør, som det ikke er bare uttrykt i MSC men også i lipofibroblasts 5,20. I denne protokoll har vi eksplisitt valgt en på magnetiske kuler valg, som det er mildere på cellene, og hele prosedyren kan utføres i sterile betingelser. En annen viktig fordel med denne isolasjonsmetoden i motsetning til den utvekst metode, er at det er forholdsvis rask, 6-10 dager i motsetning til en måned eller mer for utveksten metode. Tre til fem dager etter den første isolering av mesenchymale befolkningen er klar for CD146 + sele<html> Dette skjer; etter ytterligere tre til fem dager CD146 + cellene er klare til å bli brukt til eksperimenter eller kan bli frosset for senere bruk. Den reduserte tiden av kultur forbedrer kvaliteten på cellene som MSC transdifferentiate mot fibroblaster i forlenget ex vivo kultur 19. Til slutt, på grunn av naturen av protokollen, er det mulig å anvende denne fremgangsmåte for andre arter ved ganske enkelt å velge egnede antistoffer, eller til og med til andre organsystemer ved å justere valg av fordøyelsesenzymer og inkubasjonstid. Mesenchymale stromale celler (MSC) er i økende grad anerkjent for deres terapeutiske potensial i et bredt spekter av sykdommer, inkludert lungesykdommer. I tillegg til bruk av benmarg og navlestreng MSC for eksogene celleterapi, er det også økende interesse for reparasjon og regenerativ potensialet bosatt vev MSC. Dessuten har de sannsynligvis ha en rolle i normal organutvikling og er blitt tilskrevet roller i sykdommer, spesielt de med en fibrotisk natur. Den største hinder for studiet av disse hørende vev MSCer er mangelen på en klar markør for isolering og identifisering av disse cellene. Isolasjon teknikken beskrevet her gjelder flere kjennetegn ved lunge bosatt MSC (L-MSC). Etter ofring av rottene blir lunger ble fjernet og skyllet flere ganger for å fjerne blod. Etter mekanisk dissosiasjon av skalpellen, er lungene spaltet i 2-3 timer ved bruk av en blanding av kollagenase type I, nøytral protease og DNase type I. Det som ble oppnåddd enkeltcelle-suspensjon ble deretter vasket og lagt over tettshetsgradient medium (densitet 1,073 g / ml). Etter sentrifugering blir cellene fra den inter vasket og sådd ut i kultur-behandlede kolber. Celler dyrkes i 4-7 dager i fysiologisk 5% O2, 5% CO2-forhold.
