an efficient and high yield method for isolation of mouse dendritic cell subsets

Dendritische Zellen (DCs) sind professionelle Antigen-präsentierende Zellen, in erster Linie verantwortlich für die Erfassung, Verarbeitung und Antigenen auf Antigen-präsentierenden Molekülen präsentiert T-Zellen vermittelte Immunität zu initiieren. Dendritische Zellen können in mehrere phänotypisch und funktionell heterogenen Untergruppen getrennt werden. Drei wichtige Untergruppen von Milz- dendritischen Zellen sind plasmazytoiden, CD8a Pos und CD8a Neg Zellen. Die plasmazytoiden DCs sind natürliche Produzenten von Typ I Interferon und sind wichtig für die anti-virale T-Zell-Immunität. Die CD8a Neg DC Teilmenge wird für MHC - Klasse - II - Antigen - Präsentation spezialisiert und ist in Priming CD4 - T - Zellen zentral beteiligt. Die CD8a Pos DCs sind in erster Linie verantwortlich für die Kreuzpräsentation von exogenen Antigenen und CD8 T - Zellen - Priming. Die CD8a Pos DCs nachgewiesen wurden bei der Präsentation von Glycolipid - Antigenen durch CD1d Moleküle an eine spezialisierte T cel am effizientestenl Bevölkerung als unveränderliche natürliche Killer-T (iNKT) Zellen bekannt. Verabreichung von Flt-3-Ligand erhöht die Frequenz der Migration der dendritischen Zellvorläuferzellen aus Knochenmark, letztendlich in Expansion der dendritischen Zellen in peripheren lymphoiden Organen in murinen Modellen resultiert. Wir haben dieses Modell angepasst für eine große Anzahl von funktionellen dendritischen Zellen zu reinigen , die Verwendung in der Zellübertragung Experimente in vivo Fähigkeits verschiedener DC Untergruppen zu vergleichen.

Die Ergebnisse dieses Verfahrens beruht auf der Expansion des DC Untergruppen durch murine Flt-3L durch die implantierten Melanomzellen exprimiert wird. Der B16.Flt3L Tumor wurde aus einer C57BL / 6-Maus abgeleitet und sollte mit diesem Stamm Hintergrund, um in Tiere implantiert werden Versagen des Tumors zu vermeiden wegen Ablehnung zu etablieren.
