<html> Het hart is het eerste orgaan te vormen en functioneel in het embryo worden. Het hart is opgebouwd uit vele cellijnen die voortkomen uit de eerste en tweede embryonale hart velden 1. Uit de post-bevruchting blastocyst stadium tot aan de gevormde hart, embryonale cellen moeten dus veel mobiele beslissingen over het lot te maken. Gene transcriptie wordt geregeld in een tijd- en ruimte-afhankelijke manier en is een belangrijk biologisch proces dat lot van de cel beslissing ten grondslag ligt tijdens de embryonale ontwikkeling. Een dergelijke werkwijze wordt gemedieerd door specifieke transcriptiefactoren die regelende gebieden in het genoom waaronder enhancers en promoters van cardiale genen constitutieve binden. DNA wordt rond histonen die onderhevig zijn aan modificaties zoals acetylatie, methylatie, ubiquitinylation en / of fosforylering. Histon modificatie leidt tot onderdrukte, geactiveerd of evenwichtig gentranscriptie afhankelijk van welke lysinerest van histon is gemodificeerd 2. jove_content &quot;&gt; Chromatine immunoprecipitatie assay (chip) is opgericht jaar geleden 3 en is momenteel de meest algemeen gebruikte technologie om targets van een van beide gemodificeerde histonen of transcriptiefactoren 4. Specifiek gen transcriptie is een belangrijk biologisch proces dat lot van de cel beslissing ten grondslag ligt tijdens de embryonale ontwikkeling. Het biologisch proces wordt gemedieerd door transcriptiefactoren die genomische regulatoire gebieden zoals enhancers en promoters van cardiale genen constitutieve binden. DNA wordt rond histonen die worden blootgesteld aan chemische modificaties. Modificaties van histonen verder leiden tot een onderdrukte, geactiveerd of poised gentranscriptie, dus brengt een ander niveau van fine tuning regulatie van gentranscriptie. Embryonale stamcellen (ES-cellen) herhalen binnen embryo organen (dwz cel aggregaten) of 2D kweek de vroege stappen van ontwikkeling van het hart. Ze bieden in principe genoeg materiaal voor chromatine immunoprecipitatie (chip), een technologie die in grote lijnen gebruikt om gen regulerende gebieden te identificeren. Verder menselijke embryonale cellen vormen een menselijke cel model van cardiogenese. In latere stadia van ontwikkeling, muis embryonale weefsels mogelijk makenonderzoek naar specifieke epigenetische landschappen die nodig zijn voor de bepaling van de cel identiteit. Hierin beschrijven we de protocollen van de chip, sequentiële ChIP gevolgd door PCR of ChIP-sequencing met behulp van ES-cellen, embryo lichamen en cardiale specifieke embryonale regio&#39;s. Deze protocollen laten onderzoeken van de epigenetische regulatie van cardiale gen-transcriptie.
