La fistule aortovenous murin (FAV) modèle de ponction entre l&#39;aorte et la veine cave inférieure (VCI) est maintenant une technique bien établie. 1 Dans ce modèle, les deux parois de l&#39;aorte sous-rénale sont perforés avec une aiguille G 25, en sortant dans la attenante infra-rénale de la veine cave; le trou d&#39;entrée aortique antérieure est réparé avec la compression simple, et ne nécessite pas de suture réparation. La délivrance d&#39;agents thérapeutiques à des récipients peut être remise par l&#39;intermédiaire endovasculaire à la lumière ou par livraison externe à l&#39;adventice. Un exemple de livraison externe est la comcouramment utilisé l&#39;application adventice de gel Pluronic. Ce copolymère est thermoréversible et transformé de l&#39;état liquide à un gel solide lorsqu&#39;elle est chauffée à la température du corps. Délivrance de médicament prolongée d&#39; études antérieures ont montré est obtenu lorsque médicament mélangé dans un gel Pluronic est appliqué par voie topique in vivo. 3,4 adventiciel demande de vecteurs viraux ou d&#39; ARNsi avec un gel Pluronic a été rapporté pour être efficace en tant que système de délivrance périvasculaire 5,6 . intraluminal drug delivery to the mouse arteriovenous fistula endothelium

La délivrance d&#39;agents thérapeutiques pour améliorer la fistule artério-veineuse (FAV), la maturation peut être administré par des voies intraluminaux ou externes. Le modèle simple FAV murin a été combinée avec l&#39;administration intraluminale de solution de médicament à l&#39;endothélium veineux en même temps que la création de la fistule. Les aspects techniques de ce modèle sont discutées. Sous anesthésie générale, une incision abdominale est réalisée et l&#39;aorte et la veine cave inférieure (VCI) sont exposés. L&#39;aorte infra-rénale et IVC sont disséqués pour le serrage. Après serrage proximale et distale, le site de ponction est exposée et une aiguille G 25 est utilisé pour percer les parois de l&#39;aorte et dans la veine cave inférieure. Immédiatement après la ponction, un gène exprimant journaliste vecteur viral a été perfusé dans la VCI par la même aiguille, suivi de 15 min d&#39;incubation. Le procédé d&#39;administration virale livraison intraluminale activée plus robuste du gène à l&#39;endothélium veineux par rapport à l&#39;administration par voie externe. Cette novel méthode de livraison facilitera les études qui explorent le rôle de l&#39;endothélium dans FAV maturation et permettent l&#39;administration de médicaments intraluminal au moment de l&#39;intervention chirurgicale.
