El método de perfusión hepática que aquí se presenta permite una recolección selectiva y el estudio de la población única de marginando leucocitos hepáticas. Células hepáticas NK, también llamadas células de pozo 3, constituyen una población de células NK distintivo que residen en los sinusoides hepáticos. Los sinusoides hepáticos contienen numerosos subtipos de leucocitos de diversas actividades inmunes críticos para el organismo. Por ejemplo, las células marginando hepática (MH) asesinas naturales (NK), también conocidas como células de pozo, se caracterizan morfológicamente como linfocitos granulares grandes (LGLs) y funcionalmente como leucocitos con capacidad citotóxica espontánea, lo que permite hepática resistencia contra el establecimiento de sangre metástasis tumorales Borne. El objetivo del método presentado en este documento es para permitir la recolección selectiva de leucocitos MH, con el fin de estudiar esta población celular importante y única (y el compartimento inmune), y para dilucidar el impacto de diversas manipulaciones (por ejemplo, la activación inmune) en estas células específicas. A pesar del fracaso de la inmunidad para eliminar un tumor primario en desarrollo, la evidencia en pacientes con cáncer y en modelos animales indican que el sistema inmune puede controlar las células tumorales, las micrometástasis, y la enfermedad residual throu circulantela inmunidad mediada por células GH (CMI). Sin embargo, existe una inconsistencia aparente entre estos en capacidades in vivo y en estudios in vitro en seres humanos y en animales, que demuestran que la mayoría de las células tumorales autólogas son resistentes a la citotoxicidad mediante la circulación o leucocitos esplénicos 1,2. Esta inconsistencia se puede atribuir, al menos en parte, a la existencia in vivo de una subpoblación de leucocitos distinto, a saber, los (sinusoides) leucocitos marginando-hepática y su subpoblación de células NK activadas, es decir, células de pozo 3. De hecho, datos no publicados de nuestro laboratorio indican que en las ratas F344 células tumorales singénicas (MADB106), que se encontró resistente a circulantes y esplénicas leucocitos, fueron lisadas por células NK MH-4. Así, las células tumorales que son supuestamente &quot;NK-resistente&quot; a los leucocitos circulantes pueden ser controlados por las células MH-NK. Digno de mención, en ratones la actividad mejorada de MH-NK es evidente sólo después in vivo inmuneestimulación (por ejemplo, mediante el uso de Poly I: C o CpG-C) 5. Las células NK específicos del hígado (MH-NK) están situados dentro de la luz sinusoidales, la adhesión a las células endoteliales y células de Kupffer. Células MH-NK se caracterizan exclusivamente por gránulos densos esféricas y vesículas de varilla de núcleo 6, que contienen fosfatasa ácida como enzimas lisosomales y perforina y granzimas como sustancias bioactivas 7,8. En comparación con las células NK circulantes, las células MH-NK exhiben un mayor número y tamaño de los gránulos y vesículas 9-11. Bajo condiciones inflamatorias, se demostró que las células MH-NK a exhibir una mayor expresión de LFA-1 12, en comparación con las células NK circulantes. selective harvesting of marginating-hepatic leukocytes

Marginating-hepatic (MH) leukocytes (leukocytes adhering to the sinusoids of the liver), were shown to exhibit unique composition and characteristics compared to leukocytes of other immune compartments. Specifically, evidence suggests a distinct pro- and anti-inflammatory profile of the MH-leukocyte population and higher cytotoxicity of liver-specific NK cells (namely, pit cells) compared to circulating or splenic immunocytes in both mice and rats. The method presented herein enables selective harvesting of MH leukocytes by forced perfusion of the liver in mice and rats. In contrast to other methods used to extract liver-leukocytes, including tissue grinding and biological degradation, this method exclusively yields leukocytes from the liver sinusoids, uncontaminated by cells from other liver compartments. In addition, the perfusion technique better preserves the integrity and the physiological milieu of MH leukocytes, sparing known physiological responses to tissue processing. As many circulating malignant cells and infected cells are detained while passing through the liver sinusoids, physically interacting with endothelial cells and resident leukocytes, the unique MH leukocyte population is strategically located to interact, identify, and react towards aberrant circulating cells. Thus, selective harvesting of MH-leukocytes and their study under various conditions may advance our understanding of the biological and clinical significance of MH leukocytes, specifically with respect to circulating aberrant cells and liver-related diseases and cancer metastases.
