En este artículo se describe un protocolo detallado de formación sobre la prueba del cilindro y la escalera. La estimulación cerebral profunda del núcleo subtalámico (STN) es una opción de tratamiento eficaz para los trastornos de la enfermedad 1 y otro movimiento de Parkinson. Los mecanismos subyacentes son aún poco conocidos y multifactorial, pero una característica clave es la modulación de la actividad de la red neuronal por despolarización repetitiva de los axones en la proximidad del electrodo estimulante 2-4. Se requiere de alta frecuencia (&gt; 100 Hz) la estimulación de un efecto beneficioso en la mayoría de los objetivos del cerebro y para la mayoría de las indicaciones de DBS. efectos secundarios de la estimulación cerebral profunda de co-activación involuntaria de otras fibras, que están cubiertos por el volumen de estimulación y que están al servicio diferentes funciones, tales como el tracto piramidal. Por lo tanto, sería deseable desarrollar parámetros de estimulación, que activan preferentemente elementos neurales beneficiosos, evitando al mismo tiempo la coactivación de elementos de efectos secundarios 5,6. Aunque la neurofisiología puede ofrecer tales tuni finaOpciones ng de DBS, el progreso científico ha sido mínima durante las últimas dos décadas, debido a las estrategias de programación principalmente han sido evaluados por &quot;ensayo y error&quot; en los pacientes y restringida por las opciones de programación limitadas de dispositivos de DBS disponibles en el mercado, en lugar de utilizar una visión neurofisiológica y define parámetros experimentales para explorar sistemáticamente el espacio de parámetros completa. Para superar el obstáculo de la traducción en la investigación DBS que proponemos un protocolo para detectar parámetros de estimulación alternativas en modelos de roedores de Parkinson antes de la exploración clínica. Enfermedad unilateral de Parkinson en ratas se modela utilizando inyecciones de 6-hidroxidopamina en el derecho medial del cerebro anterior paquete de 7,8. La lesión resultante, como se describe con más detalle hemiparkinsonianas, se evalúa en el ensayo de apomorfina por la evaluación de la puntuación de la rotación después de la inyección de apomorfina a dosis bajas y confirmada post mortem por tirosina hidroxilasa immunohistochemistry. El método es fácil de aplicar y altamente reproducible, teniendo una baja mortalidad y morbilidad. Los déficits motores resultantes son muy discretas 7,8; los animales presentan un ligero deterioro de la pata contralateral izquierdo durante tanto la exploración espontánea y agarre comportamiento complejo 9,10. Para evaluar la eficacia de los protocolos de estimulación cerebral profunda se requieren pruebas que permiten medir un cambio rápido y confiable en el rendimiento del motor y se pueden repetir en el tiempo con diferentes ajustes de neuroestimulación. Varios grupos han propuesto diferentes enfoques de estimulación y diferentes pruebas para evaluar las funciones motoras en ratas 11 con resultados muy variables e inconsistentes 11-14. Esto nos obligó a elegir un conjunto de pruebas con alta predecir la validez y la complementariedad. Además, para la evaluación del resultado motor en condiciones de estimulación cerebral profunda, fueron favorecidos pruebas de que podría ser realizado por aniani- conectado mediante un cable al generador de estímulo. A estos efectos establecimos nuestra batería de pruebas que consiste en una prueba para el uso de la pata asimetría y una prueba para alcanzar experto. El diseño del estudio se ilustra en la Figura 1. Para el uso de la pata espontánea se realizó la prueba del cilindro descrito por Shallert 15, que es una prueba ampliamente utilizado para el uso de la pata durante la exploración vertical. No se requiere ninguna preparación de cada animal. Para la evaluación del comportamiento de agarre más complejo establecimos el ensayo de la escalera de acuerdo con Montoya 16. Nuestro protocolo se modifica de acuerdo a Kloth 17. Las ratas son entrenados por un período de doce días para llegar a gránulos de la caja de la prueba. Después del período de formación de la prueba se puede aplicar para medir el comportamiento de agarre complejo contando la tasa de éxito se describe como el número de gránulos comidos. La estimulación cerebral profunda del núcleo subtalámico es una opción de tratamiento eficaz para la enfermedad de Parkinson. En nuestro laboratorio hemos establecido un protocolo para detectar distintos patrones de neuroestimulación en hemiparkinsonianas (unilateral) ratas lesionadas. Consiste en crear la lesión de un Parkinson unilateral mediante la inyección de 6-hidroxidopamina (6-OHDA) en la derecha haz medial del cerebro anterior, la implantación de electrodos de estimulación crónicas en el núcleo subtalámico y la evaluación de los resultados motores al final de 24 periodos hr de neuroestimulación externa cable unido . La estimulación se llevó a cabo con la estimulación de corriente constante. La amplitud se estableció un 20% por debajo del umbral individual para los efectos secundarios. La evaluación de los resultados del motor se realiza mediante la evaluación de la utilización de la pata espontánea en el ensayo de cilindro de acuerdo con Shallert y por la evaluación de alcanzar el experto en la escalera de prueba de acuerdo a Montoya. Este protocolo se describe en detalle la formación en la caja de escalera, el cprueba ylinder, así como el uso tanto en ratas hemiparkinsonianas. El uso de ambas pruebas es necesario, porque el ensayo de la escalera parece ser más sensible para afectar destrezas motoras finas y presenta una mayor sensibilidad a los cambios durante la neuroestimulación. La combinación del modelo unilateral Parkinson y las dos pruebas de comportamiento permite la evaluación de los diferentes parámetros de estimulación de una manera estandarizada.

Todos los animales fueron sometidos a una autopsia verificación histológica de la lesión dopaminérgica tanto y la ubicación de los electrodos. Deep brain stimulation (DBS) is an effective treatment option for Parkinson's disease. We established a study design to screen novel stimulation paradigms in rats.
