Gliaceller, som är de mest förekommande celler i det centrala nervsystemet (CNS), består av oligodendrocyter (de myeliniserande cellerna i CNS), oligodendrocyter prekursorer (OP, även känd som &quot;NG2 celler&quot;), astrocyter och mikroglia. Det finns ett växande intresse för funktioner gliaceller och deras potentiella roll i neurologiska sjukdomar 1. Till exempel är Canavan sjukdom (CD) en ärftlig neurodegenerativ sjukdom börjar tidigt i barndomen med spong leukodystrofi och en progressiv förlust av nervceller, vilket leder till döden vanligen före 10 års ålder 2,3. Mutationer i Aspartoacyclase (ASPA) gen som leder till drastiskt minskad ASPA aktivitet 4 i CD har identifierats. På cellnivå, analyser för enzymatisk aktivitet liksom in situ hybridisering (ISH) och immunohistokemi (IHC) analyser tyder på att ASPA huvudsakligen uttrycks i oligodendrocyter i hjärnan men inte i neuroner eller astrocyter 11-16. Några studier har visat att ASPA kan ocksåuttryckas i mikroglia i CNS 12,14. Hittills uppgifter om ASPA uttryck i OPS är begränsade. Enligt en färsk studie där transcriptomes av olika celltyper i musen hjärnbarken inklusive neuroner, astrocyter, OP, nybildade oligodendrocyter, myeliniserande oligodendrocyter, mikroglia, endotelceller och pericyter analyserades med RNA-sekvensering 17, är ASPA uteslutande uttrycks i oligodendrocyter , i synnerhet i myeliniserande oligodendrocyter (http://web.stanford.edu/group/barres_lab/brain_rnaseq.html). Trots dessa studier ASPA uttrycksmönster i hjärnan, ett antal osäkerheter kvarstår. Olika tekniker kan användas för att studera gen-uttrycksmönster. Det kan dock vara tekniskt utmanande att använda flera prober samtidigt för att lokalisera en genuttryck till specifika celltyper. combining double fluorescence in situ hybridization with immunolabelling for detection of the expression of three genes in mouse brain sections

Detektion av genuttryck i olika typer av hjärnceller, t.ex., neuroner, astrocyter, oligodendrocyter, oligodendrocyt prekursorer och mikroglia, kan hämmas av bristen på specifika primära eller sekundära antikroppar för immunfärgning. Här beskriver vi ett protokoll för att detektera uttrycket av tre olika gener i samma hjärnavsnittet under användning dubbel fluorescens in situ hybridisering med två genspecifika prober, följt av immunfärgning med en antikropp med hög specificitet riktad mot det protein som kodas av en tredje gen. Den Aspartoacyclase (ASPA) genen, mutationer av vilket kan leda till en sällsynt mänsklig vit substans sjukdom - Canavan sjukdom - tros uttryckas i oligodendrocyter och mikroglia men inte i astrocyter och neuroner. Men den exakta uttrycksmönstret av ASPA i hjärnan har ännu inte fastställts. Detta protokoll har tillåtit oss att bestämma att ASPA uttrycks i en delmängd av mogna oligodendrocyter ennd den kan i allmänhet tillämpas på ett brett spektrum av genuttryck mönsterstudier.
