Het doel van deze studie was om bestaande celisolatie protocollen aan te passen aan de opbrengst van coronaire gladde van muizen harten verhogen. Meeste pionierswerk in het gladde spierweefsel biologie werd uitgevoerd met gekweekte ratten aorta gladde spiercellen. Deze studies voorzien fundamentele kennis van de moleculaire mechanismen die VSMC groei, migratie en hypertrofie 7 te controleren. Aangezien het gebied vorderde, bleek dat VSMC fenotype en functie werd gecontroleerd door een aantal factoren vaatbed....

Het doel van deze methode is het vasculaire gladde spiercellen (VSMC&#39;s) van de muizen coronaire circulatie te isoleren voor gebruik in celkweek en standaard celkweek assays. We ontwikkelden deze techniek om de moleculaire mechanismen van vasculaire remodellering bij diabetes beoordelen. We hebben eerder gemeld naar binnen hypertrofische remodeling in het septum coronaire arteriolen in de db / db muismodel van diabetes 1. Vanwege de beperkte hoeveelheid weefsel in de muis septale kransslagaders standaard experimentele technieken onderzoeken eiwit veranderingen (bijvoorbeeld western blot) in db / db muizen en controlemuizen zijn best moeilijk. Bovendien hebben we eerder aangetoond dat de angiotensine receptor blokker (ARB) losartan vermindert de herinrichting waargenomen bij db / db muizen 2. Daarom, isolatie van de primaire VSMC&#39;s van de coronaire circulatie stelt ons in staat om verder te onderzoeken veranderingen in VSMC fenotype of geactiveerde signaalwegen in diabetische muizen, die contribu kan zijniten op nadelige coronaire arteriole remodeling. Talrijke studies hebben canonieke signaalwegen met behulp van VSMC&#39;s geïsoleerd van knaagdieren aorta, toegelicht in plaats van in elke specifieke vasculaire bed. Wij hebben echter aangetoond vasculaire bed specifieke remodeling in coronaire aorta en mesenterische circulatie van db / db-muizen 1, suggereert de VSMC&#39;s in elk vaatbed kunnen verschillen. Daarom is het noodzakelijk om VSMC van elkaar vaatbed isoleren teneinde beter de pathologische veranderingen in elke set VSMC. Er zijn een groot aantal verschillende methoden voor het isoleren en kweken van aorta VSMCs. Echter, nu is er slechts één studie die op de isolatie van VSMC&#39;s van de muis coronaire circulatie 3 is gepubliceerd. Teng et al. als eerste een werkwijze voor het isoleren rapporteren van VSMC&#39;s de muis coronaire circulatie; Maar we hebben het protocol ingrijpend gewijzigd omdat ze ook geïsoleerde endotheliale cells. Andere labs hebben ook gebruik gemaakt van het protocol van Teng et al. Coronaire arteriële myocyten en luchtwegen spiercellen 4,5 gladde isoleren. De veranderingen die we hebben opgenomen zullen een populatie cellen sterk verrijkt voor VSMC&#39;s van de coronaire circulatie verkregen. De retrograde perfusie van de geïsoleerde zoogdieren hart, of Langendorff techniek, werd opgericht in 1897 6 van Oscar Langendorff en wordt nog steeds veel gebruikt vandaag de dag voor de isolatie van cardiovasculaire cellen. De techniek die hier, in combinatie met de vooruitgang van de moderne murine genetische modificaties, een waardevol instrument voor nader onderzoek van de moleculaire gedrag van VSMC&#39;s van de coronaire circulatie.

Terwijl de isolatie en kweek van vasculaire gladde spiercellen (VSMC&#39;s) van grote recipiënten inmiddels goed, zochten wij isoleren en kweken van VSMC&#39;s de coronaire circulatie. Harten met intacte aorta bogen werden verwijderd en geperfuseerd via retrograde Langendorff spijsvertering oplossing die 300 eenheden / ml collagenase type II, 0,1 mg / ml sojaboon-trypsineremmer en 1 M CaCl2. De perfusates werden verzameld bij 15 minuten intervallen gedurende 90 min, tot pellet gecentrifugeerd, geresuspendeerd in uitplaatmedia en geplaat op weefselkweekplaten. VSMC&#39;s werden gekenmerkt door de aanwezigheid van SM22α, α-SMA en vimentine. Een van de belangrijkste voordelen van deze techniek is de mogelijkheid om VSMC isoleren van de coronaire circulatie van muizen. Hoewel het kleine aantal cellen verkregen kunnen sommige van de toepassingen waarvoor de cellen kan worden gebruikt te beperken, kan geïsoleerd coronaire VSMC&#39;s worden gebruikt in een verscheidenheid aan goed gevestigde celkweektechnieken en ASSAys. Studies die VSMC&#39;s van genetisch gemodificeerde muizen kan verdere informatie over de structuur-functie en signalering processen die verband houden met vasculaire pathologieën te bieden.

Door de nieuw aspect van onze coronaire VSMC isolatietechniek wilden we de zuiverheid van de celisolatie bepalen. Muis coronaire VSMC&#39;s werden geïdentificeerd op basis van hun morfologie en immunofluorescentie kleuring tot passage 2.
