De mest brukte endotelceller i forskning er menneskelige navlevene endotelceller (HUVECs) på grunn av deres bekvemmelighet og enkel dyrking. Imidlertid er en rekke undersøkelser krever tilgjengelighet av en fysiologisk modell oppnådd ved isolering av endotelceller fra andre spesifikke organer 10. Musemodeller av forskjellige kardiovaskulære sykdommer og metabolske forstyrrelser bærer fysiologiske og molekylære forandringer som er tilsvarende de som finnes hos pasienter. Videre genetisk endring av mus er et kraftig verktøy som gjør det mulig for oss å undersøke patogen rolle spesifikke gener i utvikling og progresjon av sykdommer. I dag, celle-type-spesifikke gen-knock-in eller utsjekking mus enkelt ut i mange laboratorier og måling av mRNA og proteinnivåer i spesifikke celletyper er det første skrittet i å avgjøre om musene ble virkelig genmodifisert. Endotel er et tynt enkelt lag av endoteliale celler (ECS) som linjer indre vaskulære veggen. Endoteliale celler spiller en viktig rolle ved regulering av vaskulær spenning, vaskulær permeabilitet, vaskulær og ny formasjon 1,2. Endotelial dysfunksjon er et kjennetegn på mange patologiske tilstander og endringer i endotelfunksjon kan føre til ulike bilendiovascular sykdommer 3,4. Det er således viktig å studere funksjonen av endotelceller under fysiologiske og patofysiologiske betingelser. Det er flere måter å isolere ECS 5-8 og isoleringsmetoden må optimaliseres avhengig av hvilken vev og arter vil bli brukt. Dette er fordi ECS fra forskjellige vev vise en høy grad av heterogenitet med hensyn til sine overflatemarkører og proteinekspresjon 9. Vellykket isolering av endotelceller kan ofte være utfordrende og krever en viss grad av opplæring og praksis. Når oppnådd, fremgangsmåten for isolering foreslått i denne protokollen viser seg å være stabil og effektiv. Det overordnede målet med denne metoden er å få mus koronar egenkapitalbevis (MCECs) av høy kvalitet og kvantitet. Selv om mengden av celler oppsamlet fra en hjerte er mindre enn celler oppsamlet fra andre vev ved hjelp av andre teknikker, gir denne teknikken likevel better kvalitet. Renheten av endoteliale celler i det resulterende populasjonen er større enn 90%. Dermed vil denne teknikken være ideelt for applikasjoner som er avhengige av rent isolerte endotelceller som celle bildebehandling.

Endotelceller linje innerveggen av blodkar og spiller en viktig rolle ved regulering av vaskulær tone, vaskulær permeabilitet, vaskulær og ny formasjon. Endotelcelle dysfunksjon er implisert i utvikling og progresjon av mange kardiovaskulære sykdommer, inkludert iskemisk hjertesykdom. For å undersøke funksjonen og karakterisering av koronare endoteliale celler, er celleisolasjon det første trinnet og det kreves høy renhet og antall for å gjennomføre etterfølgende eksperimenter. Denne protokollen beskriver en effektiv metode for å isolere voksen muse koronare endoteliale celler. Musen hjerte dissekeres og hakket opp i små biter. Etter fordøyelse av hjertet ved hjelp av dispase og kollagenase II, blir cellene vasket og inkubert med magnetiske kuler som er konjugert med anti-CD31-antistoff. Perlene med endotelceller vaskes flere ganger og er klar til bruk i ulike applikasjoner, inkludert bildebehandling og molekylær biologisk experiments. Effektiv isolasjon gir ca 10 4 celler per ett hjerte med over 90% renhet.

Den vellykkede isolering av koronar endotelceller fra en mus hjerte gir vanligvis rundt 10 4 celler med over 90% renhet med en brostein form. For en ren populasjon av endotelceller, bør man vise forsiktighet gjennom hele protokollen for å sikre et sterilt miljø smuss.
