Tröpfchen Mikrofluidik eine Plattform für einen hohen Durchsatz biology von 1 in einem Trägeröl suspendiert Experimente in einer Vielzahl von wässrigen Tröpfchen Kompartimentierung. Droplets sind für Anwendungen als Einzelzellanalyse 2, digitale Polymerase - Kettenreaktion (PCR) 3, wie variiert verwendet worden und Enzym Evolution 4. Fluoreszierende Assays sind die Standard-Erfassungsmodus für Tröpfchen Mikrofluidik, als ihre helle Signale und schnelle Zeitverhalten kompatibel sind mit Sub-Nanoliter Tropfenvolumen bei Kilohertz Raten zu erfassen. Viele Anwendungen erfordern Fluoreszenzdetektion, um gleichzeitig mindestens zwei Farben. Zum Beispiel unserem Labor führt häufig Tröpfchensortierexperimente-PCR aktiviert , die ein Detektionskanal für das Ergebnis eines Assays zu verwenden, und verwendet eine Sekundärhintergrundfarbstoff Assay negative Tröpfchen zählbaren 5 zu machen. Typische Nachweisstationen für Tröpfchen Mikrofluidik sind based auf Epifluoreszenz- Mikroskope und erfordern Licht Manipulationen Systeme kompliziert Anregungslicht von freien Raum Laser in Mikroskop eingeführt werden, die Probe fokussiert werden. Nach Fluoreszenz von einem Tröpfchen abgegeben wird, wird das emittierte fluoreszierte Licht gefiltert, so daß jeder Detektionskanal eine Photovervielfacherröhre (PMT) auf einem Wellenlängenband zentriert verwendet. Epifluoreszenzmikroskop basierte optische Detektionssysteme bieten eine Zutrittsschranke aufgrund ihrer Kosten, die Komplexität und die erforderliche Wartung. Optische Fasern liefern die Mittel eine vereinfachte und robuste Detektionsschema zu konstruieren, da Fasern manuell in mikrofluidischen Vorrichtungen eingesetzt werden, wodurch die Notwendigkeit für spiegelbasierte Lichtführung zu entfernen und so dass Lichtwege unter Verwendung von Lichtwellenleiterverbindern angeschlossen zu werden. In diesem Dokument beschreiben wir die Montage und Validierung eines kompakten und modularen Schema Mehrfarbenfluoreszenzdetektion durchzuführen, indem eine Anordnung von optischen Fasern eine Verwendungda einzelne Photodetektor 6. Lichtleitfasern sind mit individuellen Lasern und eingesetzt sind senkrecht zu einer L-förmigen Strömungskanal in regelmäßigen räumlichen Offsets. Ein Fluoreszenzsammelfaser wird an die Anregungsbereiche parallel orientiert und an einem einzigen PMT verbunden. Weil ein Tröpfchen zu unterschiedlichen Zeiten den Laserstrahlen durchläuft, durch den PMT aufgezeichneten Daten zeigt einen zeitlichen Versatz, dass der Benutzer zwischen der emittierten Fluoreszenz zu unterscheiden erlaubt, nachdem die Tröpfchen durch jede einzelne Laserstrahl angeregt wird. Diese zeitliche Verschiebung eliminiert die Notwendigkeit emittierte Licht zu trennen PMTs zu trennen, um eine Reihe von dichroitische Spiegel und Bandpassfilter verwendet wird. Um die Wirksamkeit des Detektors zu validieren, zu quantifizieren wir Fluoreszenz in Tröpfchenpopulationen Farbstoffe unterschiedlicher Farbe und Konzentration einkapselt. Die Empfindlichkeit des Systems wird für einfarbige Fluorescein Detektion untersucht und zeigt die Fähigkeit Tröpfchen mit Konzentrationen zu detektieren bis zu 0,1 nm, eine 200x Empfindlichkeit improvement im Vergleich zu bisherigen Vorgehensweisen auf Faserbasis 7 in der Literatur berichtet.

multicolor fluorescence detection for droplet microfluidics using optical fibers

Fluorescence assays are the most common readouts used in droplet microfluidics due to their bright signals and fast time response. Applications such as multiplex assays, enzyme evolution, and molecular biology enhanced cell sorting require the detection of two or more colors of fluorescence. Standard multicolor detection systems that couple free space lasers to epifluorescence microscopes are bulky, expensive, and difficult to maintain. In this paper, we describe a scheme to perform multicolor detection by exciting discrete regions of a microfluidic channel with lasers coupled to optical fibers. Emitted light is collected by an optical fiber coupled to a single photodetector. Because the excitation occurs at different spatial locations, the identity of emitted light can be encoded as a temporal shift, eliminating the need for more complicated light filtering schemes. The system has been used to detect droplet populations containing four unique combinations of dyes and to detect sub-nanomolar concentrations of fluorescein.
