C2C12 cellinje har använts som en modell för att studera myogenes 11-14. Dystrofia myotonika (DM) är en autosomalt dominant sjukdom som drabbar flera system, främst hjärt- och skelettmuskler 1. Det finns två subtyper av sjukdomen, DM1 och DM2. DM1 är vanligare och har en mer allvarlig manifestation än DM2 2. Den genetiska mutationen bakom DM1 är en utvidgning av CUG triplett upprepningar belägna i 3 &#39;otranslaterade regionen (UTR) av DM proteinkinas-genen (DMPK) 3. SAG upprepa nummer i opåverkade individer varierar från 5 till 37. Däremot ökar det till mer än 50, och ibland upp till tusentals i DM1 patienter 4. Som ett resultat, RNA-bindande proteiner, såsom muscleblind liknande en (MBNL1), CUGBP och Elav-liknande familjen 1 (Celf1), är misregulated. På grund av den sekvestrering på de expanderade CUG upprepningar, förlorar MBNL1 dess förmåga att reglera alternativ splitsning 5. Celf1, å andra sidan, är uppreglerad 6,7. Men modellering DM1 i myoblaster erbjuder ett effektivt alternativ, särskilt för att dissekera DM1-relaterade cellulära och molekylära händelser. C2C12 myoblast-cellinjen isolerades först från skadade C3H mus muskel och används i stor utsträckning för att studera myogenic differentiering 8,9. C2C12-celler förökar sig snabbt i fetalt bovint serum (FBS) innehållande media och lätt undergår differentiering när FBS utarmas. Men att använda denna myoblast differentiering modell presenterar två utmaningar: C2C12-celler är ofta resistenta mot gen transfektion / virusöverföring; och små variationer i cell hantering och differentiering förfarandet kan leda till påtagliga förändringar i myotube bildning. Vårt laboratorium rutinmässigt använder C2C12 myoblaster som acell modell och har utvecklat protokoll som effektivt leverera gener från plasmid transfektion, retroviral transduktion, och lentivirala transduktion i C2C12 cellinje 10. I videon visar vi optimerade förfaranden för transfektion / transduktion C2C12-celler och upprätthålla differentiering konsekvens i upprättandet DM1 myoblast modeller.

modeling myotonic dystrophy 1 in c2c12 myoblast cells

Dystrofia myotonika 1 (DM1) är en vanlig form av muskeldystrofi. Trots att flera djurmodeller har fastställts för DM1, myoblast cellmodeller är fortfarande viktiga eftersom de erbjuder ett effektivt cellulär alternativ för att studera cellulära och molekylära händelser. Fastän C2C12 myoblastceller har använts i stor utsträckning för att studera myogenes, resistens mot gen transfektion eller viral transduktion, hindrar forskningen i C2C12-celler. Här beskriver vi en optimerad protokoll som inkluderar daglig underhålls transfektion och transduktion förfaranden för att införa gener i C2C12 myoblaster och induktion av myocyt differentiering.
