La plupart des virus varient en taille de 20 à 300 nm, bien que la pandoravirus est supérieure à 700 nm de taille 1. La distribution des particules virales dans la phase aiguë de l&#39;infection peut dépendre de la taille des particules, la distribution des récepteurs cellulaires, ou l&#39;affinité des récepteurs cellulaires pour le virus. Dans les modèles animaux, intracérébroventriculaire (ICV), intrapéritonéale, placentaires directe, et par voie intraveineuse (IV) infections ont été utilisées pour étudier la pathogenèse de l&#39;encéphalite virale. Dans l&#39;étude sur la pathogenèse de l&#39;encéphalite virale, la méthode d&#39;infection est critique. La première des deux voies principales infectieuses vers le cerveau est la voie hématogène, ce qui implique l&#39;infection des cellules et les pericytes du cerveau endothéliales. Le second est le intracérébroventriculaire (ICV) itinéraire. Une fois dans le système nerveux central (SNC), les virus peuvent se propager dans l&#39;espace sous-arachnoïdien, des méninges et plexus choroïde par le liquide céphalorachidien. Dans des modèles expérimentaux, les premiers stades de la distribution virale CNS ne sont pas bien caractérisées, et il est difficile de savoir si seules certaines cellules sont initialement infectées. Ici, nous avons analysé la distribution du cytomégalovirus (CMV) particules pendant la phase aiguë de l&#39;infection, appelée virémie primaire, suivant ICV ou intravasculaire (IV) l&#39;injection dans le cerveau de souris néonatale. Dans le modèle d&#39;injection ICV, 5 pi de CMV murin (MCMV), ou des microbilles fluorescentes ont été injectées dans le ventricule latéral du midpoint entre l&#39;oreille et l&#39;oeil en utilisant une seringue de 10 ul avec une aiguille 27G. Dans le modèle d&#39;injection intraveineuse, une seringue de 1 ml avec une aiguille de 35 G a été utilisée. Un transilluminateur a été utilisé pour visualiser le temps (faciale) veine superficielle de la souris néonatale. Nous infusé 50 pi de MCMV ou microbilles fluorescentes dans la veine temporale superficielle. Les cerveaux ont été récoltés à différents moments après l&#39;injection. Génomes Mcmv ont été détectés en utilisant la méthode dans d&#39;hybridation in situ. microbilles fluorescentes ou protéine fluorescente verte exprimant des particules Mcmv recombinantes ont été observées par microscopie à fluorescence. Ces techniques peuvent être appliquées à de nombreux autres agents pathogènes pour étudier la pathogenèse de l&#39;encéphalite.
