De meest kritische stap in de oprichting van een MSPP regeling is de ondubbelzinnige genetische bepaling van biomarkers ion identiteiten. Als het niet mogelijk is om een biomarker ongetwijfeld identificeren, dan moet worden uitgesloten van de regeling 13. De C. jejuni ssp. doylei project omvat 14 verschillende biomarker ionen. Tijdens het laatste decennium, heeft-MALDI time-of-flight massaspectrometrie (MALDI-TOF MS) gevorderd tot een zeer gewaardeerd standaardmethode voor microbiële geslacht en soort identificatie in klinische microbiologie 1, 2 zijn. Soortidentificatie is gebaseerd op de registratie van kleine eiwit vingerafdrukken van intacte cellen of cellysaten. De typische massabereik voor een massaspectrometer gebruikt in routine klinische microbiologie is 2-20 kDa. Bovendien kan de resulterende spectra worden gebruikt om spanningen op de hiernavolgende-species en subspecies niveau beneden-3 discrimineren. Vroege baanbrekende studies hebben specifieke biomarker ionen geïdentificeerd voor een bepaalde subgroep van stammen in Campylobacter jejuni 4, Clostridium difficile 5, Salmonella enterica ssp. enterica serovar Typhi 6, Staphylococcus aureus 7-9 en Escherichia coli 10-12. De combinatie van verschillende variabele biomarker massa&#39;s die overeenkomen met allele isovormen biedt de mogelijkheid voor diepere subtypering. Eerder hebben we met succes een methode om deze variaties in de massa-profielen om te zetten in betekenisvolle en reproduceerbare phyloproteomic relaties genaamd massaspectrometrie gebaseerd phyloproteomics (MSPP) op een C. geïmplementeerd jejuni ssp. jejuni isolatencollectie 13. MSPP kan worden gebruik gemaakt van een massaspectrometrische gelijk aan DNA-sequentie gebaseerd subtypering technieken zoals multilocus reeks typen (MLST). Campylobacter soorten zijn de belangrijkste oorzaak van bacteriële gastroenteritis wereldwijd 14, 15. Als gevolg van Campylobacteriose post-infectueuze sequela, namelijk Guillain Barré, reactieve artritis en inflammatoire darmziekte kunnen ontstaan 16. De belangrijkste bronnen van infectiebesmet vee vlees van kip, kalkoen, varkens, runderen, schapen en eenden, melk en oppervlaktewater 15, 17. Daarom regelmatig epidemiologische surveillance studies in het kader van de voedselveiligheid zijn noodzakelijk. MLST is de &quot;gouden standaard&quot; in moleculaire typering voor Campylobacter soorten 18. Omdat de Sanger-sequencing-gebaseerde MLST werkwijze is arbeidsintensief, tijdrovend en relatief duur is MLST typen beperkt tot relatief kleine isoleren cohorten. Daarom is er behoefte aan goedkopere en snellere subtypering methoden. Deze behoefte kan worden voldaan door massaspectrometrische methoden zoals MSPP. Dit document presenteert een gedetailleerd protocol voor MSPP-typering met behulp van een verzameling van Campylobacter jejuni ssp. Doylei isolaten en vergelijking van het potentieel met MLST.

MALDI-TOF MS biedt de mogelijkheid om sommige bacteriën niet alleen op de soorten en ondersoorten niveau, maar ook onder, De stam differentiëren. De C. jejuni ssp. doylei MSPP regeling omvat 14 verschillende biomarker ionen, vooral ribosomale eiwitten in het massabereik van 2-11 kDa. MSPP kan in principe worden aangepast aan andere massaspectrometrische platforms met een uitgebreid massagebied.
