Volwassen adipocyten dat ondergaan in vitro dedifferentiatie, een proces dat bekend staat als het plafond cultuur, kan terugkeren naar een meer primitieve fenotype en krijgen proliferatieve capaciteiten. Celtherapie en tissue engineering zijn hot topics op het gebied van regeneratieve geneeskunde 1-5. Hoewel deze therapeutische modaliteiten zijn veelbelovend, een aantal technische kwesties die momenteel belemmeren hun klinisch gebruik. In dit opzicht, zoals bij het genereren van iPS cellen, alle weefselmanipulatie therapieën moeten cellen aan externe gen transductie produceren om patiëntveiligheid handhaven. Daarom waren we de eerste groep met succes de productie van menselijke cellen DFAT 6. Verscheidene andere onderzoeksgroepen hebben sindsdien aangenomen onze methode om DFAT cellen van zoogdieren oorsprong 7-9 te genereren, verder aandacht voor de bruikbaarheid van ons model. Gedurende verscheidene studies hebben we gevonden dat de kwaliteit van de celkweek omgeving kan worden gewijzigd door instellen van de inhoud van het celmedium. Deze bevinding heeft geleid tot een verhoging van de slaagkans van DFAT celproductie en cellen kwaliteit verbeterd; zowel kritische factoren inefficiënt genereren van cellen voor toekomstige klinische trials. In dit opzicht een verbeterde DFAT kweekmedium (DCM, een medium vergelijkbaar met mesenchymale stamcel medium dat recombinant humaan insuline, serum albumine, L-glutaminezuur, verscheidene vetzuren bevat, en cholesterol) en een werkwijze voor DFAT celgeneratie en proliferatie werd ontwikkeld (meer informatie over de inhoud van DCM is beschikbaar op aanvraag). Deze methode hoogwaardige DFAT cellen werden gegenereerd met de mogelijkheid om te differentiëren in verschillende celtypes waaronder adipogenic, osteogene, chondrogene en cellen. Al met al, deze gevalideerd celkweek protocol verhoogt de kwaliteit van DFAT cellen en kunnen zeer bruikbaar voor het verbeteren van klinische toepassingen van celtherapie en tissue engineering.

Tissue engineering en celtherapie veelbelovend klinisch. In dit verband kan multipotente cellen, zoals mesenchymale stamcellen (MSCs), therapeutisch worden toegepast, in de nabije toekomst de functie herstellen beschadigde organen. Toch een aantal technische kwesties, waaronder de zeer invasieve procedure voor het isoleren van MSC&#39;s en de inefficiëntie rond hun versterking, momenteel belemmeren de potentiële klinische gebruik van deze therapeutische modaliteiten. Hierin introduceren we een zeer efficiënte werkwijze voor de productie van gededifferentieerde vetcellen (DFAT), MSC-achtige cellen. Interessant is DFAT cellen worden gedifferentieerd in verschillende celtypen waaronder adipogenic, osteogene, chondrogene en cellen. Hoewel andere groepen die eerder verschillende methoden hebben ingediend voor het genereren van DFAT cellen uit volwassen vetweefsel, onze methode laat ons toe om DFAT cellen efficiënter te produceren. Hierbij tonen we dat DFAT kweekmedium (DCM), aangevuld met 20% FBS,effectiever in het genereren DFAT cellen dan DMEM, aangevuld met 20% FBS. Bovendien kan de DFAT cellen die door onze celkweek methode redifferentiated in verschillende weefseltypen. Als zodanig is een zeer interessant en nuttig model voor de studie van weefsel dedifferentiatie gepresenteerd.
